摘要
目的检测炎症性肠病(IBD)患者血清中调节性T细胞(Treg)及转化生长因子β(TGFβ)、IL-10的表达水平,探讨其在IBD中的作用。方法采用流式细胞术检测IBD患者外周血血清中Treg细胞(CD4+CD125highCD127-)的水平,ELISA法检测IBD患者血请中TGFβ、IL-10的含量,Real-time PCR检测IBD患者肠粘膜组织中TGFβ、IL-10的基因表达水平;免疫组化染色分析肠粘膜组织中TGFβ、IL-10的表达。30例正常人作为对照组。结果 IBD组患者外周血中调节性T细胞占CD4+T细胞的比率明显低于健康对照组(P<0.05),血清中TGFβ与IL-10的表达水平均明显降低(P<0.05);IBD患者肠粘膜组织中TGFβ与IL-10的mRNA水平及蛋白水平均低于正常对照组。结论 Treg及TGFβ、IL-10对于维持机体免疫平衡具有重要作用,Treg及TGFβ、IL-10的表达水平降低是IBD发病的重要机制之一。
Objective To determine the expression and significance of Treg,TGFβand IL-10 in the serum of patients with inflammatory bowl disease(IBD). Methods The level of Treg(CD4 +CD125highCD127-)in the peripheral blood was determined with flow cytometery,the content of TGFβand IL-10 in the plasma was evaluated with ELISA,the mRNA level of TGFβand IL-10 was determined with real-time PCR,and the expression of TGFβand IL-10 in intestinal mucosal tissue was detected with immunohistochemistry.50 IBD patients formed the experimental group and 30 healthy cases formed the control group.Results The number of Treg in CD4+T cells was significantly lower in the experimental group than that in the control group(P 〈0.05).The levels of TGFβand IL-10 decreased significantly in the IBD group when compared with the normal group(P 〈0.05).The expressions of TGFβand IL-10 in intestinal mucosal tissues were lower in the experimental group than those in the control group.Conclusion Treg cells,TGFβand IL-10 are important in maintaining immune balance in the body and they are involved in the development of IBD.
