阿托伐他汀对高糖诱导内皮细胞MALAT1及炎症因子表达的影响 被引量:6

Effect of atorvastatin on high glucose-stimulated expressions of MALAT1 and inflammatory factors in endothelial cells

摘要 目的探讨阿托伐他汀对高糖诱导人脐静脉内皮细胞(HUVECs)长链非编码RNAs(LncRNAs)分子转移相关肺腺癌转录物1(MALAT1)及炎症因子表达的影响。方法以实时定量PCR检测高糖(30mmol/L)刺激HUVECs不同时间后MALATl的表达水平;在高糖条件下,利用siRNA技术沉默细胞中MALATl的表达,或加阿托伐他汀处理,检测MALATl、白细胞介素(IL)-6和IL-8的表达水平。结果(1)高糖刺激HUVECs不同时间后,MALATl的表达出现了不同程度的升高(P〈0.05),在12h时MALATl表达增加最为显著(P〈0.01).高糖刺激HUVECs12h后炎症因子1L-6和IL-8的表达和分泌水平也明显升高(P〈0.05):(2)siRNA沉默MALATl表达可明显抑制高糖刺激的IL-6和IL-8表达和分泌(P〈0.05);(3)阿托伐他汀明显抑制高糖刺激的MALATl、IL-6和IL-8表达(P〈0.05)。结论高糖可通过MALAT1介导内皮细胞炎症因子的分泌,而阿托伐他汀可明显抑制高糖刺激MALAT1的表达,降低炎症反应. Objective To investigate the effect of atorvastatin on the expressions of long non-coding RNA (LncRNA) metastasis-associated lung adenocarcinoma transcript 1 ( MALAT1 ) and inflammation factors in human umbilical vein endothelial cells (HUVECs) stimulated by high glucose. Methods The expression of MALAT1 in HUVECs incubated with high glucose (30 mmol/L) for different time periods were detected by real-time PCR. Under high glucose condition, the expressions of MALAT1, interleukin-6 ( IL-6 ), and interleukin-8 ( IL-8 ) in HUVECs were detected after MALAT1 was silenced by siRNA or atorvastatin was added. Results ( l ) After HUVECs were incubated with high glucose for different time periods, the expressions of MALAT1 were increased to some extent(P〈 0.05 ) , with the peak at 12h ( P〈0.01 ). The levels of IL-6 and IL-8 expression and secretion were increased after HUVECs were stimulated by high glucose for 12h ( P〈0.05 ). ( 2 ) The silence of MALAT1 markedly suppressed high glucose-stimulated expression and secretion of IL-6 and IL-8 ( P〈0.05 ). ( 3 ) Atorvastatin significantly inhibited high glucose-stimulated expressions of MALAT1, IL-6, and IL-8 ( all P〈0.05 ). Conclusions High glucose induces the secretion of inflammatory factors by stimulating MALAT1 expression in endothelial ceils. Atorvastatin significantly inhibits high glucose-stimulated MALAT1 expression and decreases inflammatory reaction.
