摘要
目的观察牛黄参胶囊含药血清对人肝癌细胞HepG-2的Bax、Bcl-2的调控作用。方法建立并分为空白对照组,环磷酰胺组和牛黄参胶囊5%、10%和20%含药血清组。采用实时定量PCR检测Bax、Bcl-2 mRNA的表达水平;免疫组织化学法检测HepG-2细胞Bax、Bcl-2的表达;Western blotting检测Bcl-2蛋白的表达水平。结果牛黄参胶囊20%含药血清能在基因和蛋白水平明显上调Bax表达(P<0.01),明显抑制Bcl-2的表达(P<0.01),并具有明显的剂量依赖性。结论牛黄参胶囊含药血清能明显上调Bax表达,同时下调Bcl-2表达,可能是其诱导肝细胞凋亡的机制之一。
Objective To investigate the effect of the serum containing Niuhuangshen capsule （NHS） on the regu- lation of Bax and Bcl - 2 in HepG - 2. Methods The effects of the NHS on the regulation of Bax and Bcl - 2 in human hepatoma carcinoma HepG -2 were determined by real time PCR in mRNA level and the Bax/Bel -2 protein expression were evaluated by immunohistochemistry and western blotting. Results The expression of Bax was increased with dose dependent （P 〈0.01 ） in 48h, and the expression of Bcl- 2 was decreased with dose dependent（P 〈 0.01 ） in RNA and protein level. Conclusion The serum containing Niuhuangshen capsule probably induces HepG -2 apoptosis by taking advantage of up - regulation Bax and down - regulation Bcl - 2.
