籼粳交水稻花药培养条件的优化 被引量:3

摘要 旨在筛选最适出愈培养基,以水稻籼粳交品种的花药为植体,以预处理时间、2,4-D质量浓度、播期、接种密度、叶枕间距为因素的正交试验,通过方差分析研究影响籼粳交后代花药出愈率的主要因素,并对其最适值进行优化。成苗阶段研究MS培养基中加入4种不同植物激素、在不同的愈伤培养时间以及培养温度处理条件下对籼粳交材料愈伤组织成苗率的影响。结果表明,不同籼粳交材料后代均在3号M8培养基的出愈率最高;预处理时间、叶枕间距的差异达到极显著水平。在分化成苗阶段,蔗糖是影响成苗阶段的主要因素,其次为6-BA,且分别在30g/L、4mg/L时分化率最高。愈伤培养30d转接分化率最高;培养温度为29℃时绿苗率最高。表明,M8+8mg/L 2,4-D+8mg/L NAA+4mg/L KT+2mg/L 6-BA+30g/L蔗糖+6g/L琼脂为最优出愈培养基;预处理时间和叶枕间距是影响出愈率的主效因子,最适宜分化培养基配比为MS+6-BA 4mg/L+蔗糖30g/L+琼脂6g/L;最佳愈伤转接时间为30d;最佳培养温度为29℃。 In order to improve culture efficiency of anther in indica/japonica hybrid rice,cultivars of the indica/japonica hybrid rice were used for screening culture medium,the orthogonal test of five factors was designed, which the five factors were the pretreatment days, the mass concentration of 2,4-D, the sowing date,and the inoculums density and the occipital lobe spacing. Furtherly,at the seedling stage, the test based on the MS culture medium with different plant hormones, different culture time and culture temperature conditions. Seedling rate of cross material callus in indica japonica were inves- tigated in the paper. The results showed that the callus rate of three different indica/japonica hybrid materials were the highest in No. 3 M8 medium. The pre-treatment days and the inoculums density reached at very significant level. At the seedling stage, sucrose content was the primary impact factor, followed by the concentration of 6-BA. And differentiation rate of 30 g/L,4 mg/L respectively was highest. The highest differentiation rate of callus culture time was 30 d. When the culture temperature was 29 ℃ it had the highest rate of plantlet differentiation. It was concluded that M8q-8 mg/L 2,4-D+8 mg/L NAA+4 mg/L KT+2 mg/L 6-BA+30 g/L sucrose and 6 g/L agar was the optimum medium at the callus stage,while and MS+6-BA 4 mg/L+sugar 30 g/L and agar 6 g/L was the most appropriate media ratio for seedling differentiation at the seedling stage. In conclusion, the best callus transit time was 30 days and the optimum culture temperature was 29 ℃.
