摘要
目的:建立地萸补肾丸中黄芪甲苷、莫诺苷、马钱苷的含量测定方法。方法:分别采用HPLC-ELSD方法测定黄芪甲苷含量,色谱条件:Shimadzu C18色谱柱(5μm,250 mm×4.6 mm);流动相∶乙腈-水(30∶70);流速:1.0 m L·min-1;柱温:30℃;ELSD参数:漂移管温度:105℃,载气流速:2.7 m L/min;采用HPLC方法测定莫诺苷、马钱苷含量,色谱条件:Kromasil 100A C18色谱柱(5μm,250 mm×4.6 mm);流动相为乙腈(A)-0.3%磷酸溶液(B)梯度洗脱;检测波长240 nm;流速1.0 m L·min-1;柱温40℃。结果:黄芪甲苷、莫诺苷、马钱苷分别在0.4464~8.928μg(r=0.99985),0.0506~1.012μg(r=0.99997),0.0492~0.984μg(r=0.99991)范围内线性良好,平均加样回收率分别为100.18%(RSD1.25%)、99.54%(RSD1.07%)、99.60%(RSD1.60%)。结论:实验表明,该方法方便快捷、准确、灵敏度高、重现性好,可用于地萸补肾丸的质量控制。
Objective:To establish a method for content of astragaloside,morroniside and loganin in Diyu Bushen Pill. Methods:Using the HPLC-ELSD method for determining the content of astragaloside,HPLC-ELSD system consisting of a Shimadzu C18(5 μm,250 mm×4.6 mm);the mobile phase of acetonitrile water(30∶70);flow rate:1.0 m L·min-1;column temperature:30 ℃;ELSD parameters:drift tube temperature:105 ℃ and the flow rate:2.7 m L/min. Determination of morroniside and loganin by HPLC,using the Kromasil 100 A C18(5 μm,150 mm×4.6 mm);the mobile phase was acetonitrile(A)-0.3% phosphoric acid solution(B)gradient elution;the detection wavelength is at 240 nm and the flow rate was 1.0 m L·min-1;the temperature of detection wavelength column is 40 ℃. Results:Astragaloside,morroniside and loganin respectively in 0.4464-8.928 μg(r=0.99985),0.0506-1.012 μg(r=0.99997),0.0492~0.984 μg(r=0.99991)linear within the range of good and the average recoveries were 100.18%(RSD1.25%),99.54%(RSD1.07%),99.60%(RSD1.60%). Conclusion:This experiment show that the method is convenient,accurate,high sensitivity,good reproducibility and it can be used for the quality control of Diyu Bushen Pill.
