摘要
目的:研究外用中药制剂对小鼠乳腺增生的治疗作用并探讨其机制。方法:采用苯甲酸雌激素腹腔注射构建乳腺增生小鼠模型,分组为对照组、模型组与治疗组。对照组小鼠腹腔注射生理盐水,隔天1次,共30天;模型组与治疗组腹腔注射苯甲酸雌激素,隔天1次,共30天。造模完成后,治疗组小鼠乳房部涂抹外用中药制剂,每天1次共30天。测定各组小鼠体重变化,乳晕及乳头直径变化,血清雌激素(E_2)、孕酮(P)指标,并取小鼠第二对乳房组织做病理切片观察。结果:治疗组与模型组小鼠比较,体重、乳晕及乳头直径均有统计学差异(P<0.05);治疗组小鼠病理切片HE染色显示乳腺增生组织结构明显改善,雌、孕激素水平较对照组无统计学差异(P>0.05),而与模型组对比则具有统计学差异(P<0.05)。结论:外用中药制剂对小鼠乳腺增生具有改善作用,其机制可能是通过调节小鼠血清雌激素与孕酮水平来实现的。
Objective：To study and the effect of traditional Chinese medicine on the mammary gland hyperplasia in mice and to explore its mechanism.Methods：The model group was established by injecting estradiol benzoate into the abdominal cavity.The mice were divided into 3 groups（control,model and therapy）.Normal saline was injected into the peritoneal cavity of the control group,every 2 days,altogether 15 times in 30 days.Estradiol benzoate was injected into the peritoneal cavity of the model group and therapy group,every 2 days,altogether 15 times in 30 days.After 30 days,the traditional Chinese medicine was smeared on the mammary of therapy group,once a day for 30 days.To observe the body weight changes of mice,areola and nipple diameter,serum estradiol（E2）and progesterone（P）index.And the pathological slices of the second couple breast tissue were observed.Results：We found the body weight and diameter of nipple of mice had differences between therapy group and model group（P〈0.05）.The level of estrogen and progesterone compared between the therapy group and control group showed no significant difference（P〉0.05）,and compared with the model group there had statistical difference（P〈0.05）.The treatment group mice pathological HE staining showed hyperplasia of the breast tissue structure was improved.Conclusion：The improvement effect of traditional Chinese medicine preparation for mammary gland hyperplasia in mice may be realized through regulating the mouse serum estrogen and progesterone level.
