摘要
目的:采用DNA条形码技术和随机多态性(RAPD)分子标记技术对12个不同品种(系)的白芍种质资源进行遗传多样性研究。方法:采用CTAB法提取白芍叶片基因组DNA,利用ITS2引物和RAPD随机引物对样品进行PCR扩增,对于ITS2-PCR体系,测序后采用相似性搜索(BLAST法)进行鉴定分析;对于RAPD-PCR体系,根据条带数目,应用NTSYS2.10e软件采用UPGMA法进行聚类分析。结果:12个品种的ITS2序列的BLAST结果高度一致,未见碱基位点的变异。RAPD引物碱基序列共扩增出308条带,其中多态性条带有257条,多态百分率为83.4%,各品种(系)间的相似系数在0.558~0.847之间。结论:白芍的各品种间保存着丰富的遗传多样性。
Objective: To study the 12 gerplasm resources of Paeonia lactiflora Pall. by using DNA barcoding and RAPD. Methods: Genomic DNA was extracted by CATB method,ITS2 primer and RAPD primers were selected to perform PCR amplification. For ITS2-PCR amplified system,identification analysis was performed using BLAST after being sequenced. For RAPD-PCR amplified system. NTSYS-2. 10 e software was applied to the cluster analysis by UPGMA according to the band number. Results: BLAST results showed that ITS2 sequences of 12 populations were highly consistent,and no nucleotide mutation was detected. The RAPD primers had amplified 308 clear bands,257 polymorphic bands and the percentage of polymorphism was 83. 4%. The coefficient of genetic similarity of these materials ranged from 0. 558 to0. 847. Conclusion: P. lactiflora Pall. has abundant genetic diversity.
