摘要
目的探索淫羊藿素对人脐带间充质干细胞(UC-MSC)体外增殖及细胞因子分泌的影响。方法分离、培养人脐带来源的间充质干细胞(MSC),采用含不同浓度淫羊藿素的培养基培养,流式细胞术检测MSC表面标志物,Brd U法检测MSC的增殖能力,实时荧光定量PCR法检测MSC分泌细胞生长因子的变化。结果不同浓度(10,20,50,100 ng·mL^(-1))的淫羊藿素均可促进MSC增殖(P<0.05),其中以20,50 ng·mL^(-1)组促增殖作用最明显;与对照组比较,淫羊藿素对尿激酶纤溶酶原激活物(u PA,10 ng·mL-)1、基质金属蛋白酶-1(MMP-1,10,20,50 ng·mL-)1、血管内皮生长因子(VEGF,50 ng·mL^(-1))mR NA的表达明显增加,差异有统计学意义(P<0.05)。结论淫羊藿素可促进UC-MSC体外增殖,并且促进肝细胞生长因子(HGF)、u PA、MMP-1、VEGF的表达。
Objective To investigate the effects of icariin on the proliferation and secretion of cytokines of human umbilical cord mesenchymal stem cells(UC-MSC ) in vitro. Methods UC-MSC were isolated and cultured, and then were incubated with medium containing different concentrations of icaritin. We detected UC-MSC surface markers by flow cytometry, examined the proliferation ability by BrdU method, and observed the changes of UC-MSC cytokines by real-time quantitative PCR. Results Icaritin at different concentrations( 10, 20, 50, 100 ng'mL-1) could promote the proliferation of UC-MSC(P 〈 0.05), and the effect of 50 ng·mL-1 group was the most strongest. Compared with the control group, icaritin could promote the mRNA expression of urokinase type plasminogen activator (uPA; icariin in 10 ng·mL-1) , matrix metalloproteinase-1 (MMP-1; icaritin in 10, 20, 50 ng·mL-1) , and vascular endothelial growth factor (VEGF; icaritin in 50 ng·mL-1) in UC-MSC, the differences being significant(P 〈 0.05). Conclusion Icaritin can promote the proliferation of UC-MSC in vitro, and enhance the expression of cytokines (uPA, MMP-1, VEGF).
