丹红注射液促进鸡胚绒毛尿囊膜血管生成的实验研究 被引量:3

摘要 目的探讨丹红注射液对鸡绒毛尿囊膜(chick embryo chorioallantoic membrane,CAM)血管生成的影响。方法种蛋在(37.5±0.5)℃条件下孵育,第7天开窗暴露CAM建立CAM模型。将60枚存活鸡胚随机分为生理盐水组(N)、正常血清组(C)、丹红注射液治疗组(T)、血管内膜损伤组(I)、血管内膜损伤后丹红注射液治疗组(IT)以及血管内皮生成因子(vascular endothelial growth factor,VEGF)(20μg/ml)组(V),每组10枚鸡胚。新西兰大白兔腹主动脉用球囊导管损伤建立血管内膜损伤模型,丹红注射液治疗组及血管内膜损伤后丹红注射液治疗组于大白兔耳缘静脉注射丹红注射液2ml.Kg-1,各组连续7 d后取血清。第8天,在鸡胚CAM上放一直径为5 mm的滤膜作为载体,分别加样5μl,1次/d,连续3 d,第11天取CAM,数码相机拍照后平铺于载玻片上,计数载体周围血管数目及滤膜周围0.5 cm范围内的血管分支点数并进行比较。结果比较CAM周围血管数目,T组(31.80±3.08)与C组(28.40±3.75)相比较,血管以载体为中心呈辐辏状生长,血管总数明显增多,具有显著性差异;I组(47.30±3.27)与C组相比较,载体周围血管数量明显增多,具有极显著性差异;IT组(47.60±3.89)组相比较,血管总数无显著性差异。结论①含丹红注射液的兔血清能够明显促进CAM上血管生成,②血管内膜损伤后的血清和内膜损伤丹红治疗组也分别强于血管内膜未损伤时。 Objective To investigate the angiogenesis effect of Danhong injection on the chick embryo chorioallantoic membrane(CAM) model.Methods Fertilized chick eggs were incubated and made a window in every egg on day 7.60 survivors were randomly divided into 6 groups: normal saline(group N),normal rabbit serum(group N),serum of rabbit Danhong injection(group T),serum of rabbit suffered with vascular intima injury(group I),serum of Danhong injection suffered with vascular intima injury(group IT) and vascular endothelial growth factor(VEGF 20μg·ml-1)(groupV).Group T and Group IT administered Danhong injection 2 ml·kg-1 for seven days.On day 8,one disc was placed on every CAM and 5 μl sample of each group was subtracted onto discs once a day for three days.The CAMs were harvested on day 11 and the number of blood vessel around the discs was counted.Results The proliferation of vessels in group T(31.80±3.08) and group I(47.30±3.27)was obviously higher than that of group C(28.40±3.75)respectively,but compared with group I,the group IT(47.60±3.89)had no obvious difference in total number of vessels.Conclusion ①Serum of Danhong injection can stimulate angiogenesis of CAM.②The serum and Danhong injection serum from vascular intima injured rabbit has more effective angiogenesis compared to normal serum.
