以肾脑相济理论为指导的电针疗法对围绝经期抑郁症大鼠海马Notch通路的影响 被引量:18

Effect of Electroacupuncture on Expression of Hippocampal Notch Signaling Pathway in Rats with Perimenopausal Depression Guided by Theory of Kidney and Brain Communication

摘要 目的:研究以"肾脑相济"理论为指导的电针疗法对围绝经期抑郁症大鼠海马Notch通路相关指标Notch1、Jagged1、Hes1的影响,探讨以"肾脑相济"理论为指导的电针疗法对围绝经期抑郁症的治疗机制。方法:48只大鼠随机分为空白组、模型组、西药组和电针组四组,采用去势(OVX)和慢性不可预见刺激(CUMS)结合制备围绝经期抑郁症大鼠模型。西药组采用盐酸氯米帕明灌胃,电针组采用电针刺激百会、肾俞、三阴交穴。连续治疗28 d,观察大鼠一般情况变化,动情周期,强迫游泳及旷场实验结果,运用运用酶联免疫吸附法(ELISA法)检测海马组织微管相关蛋白(MAP-2)表达、ELISA法及RT-PCR法检测海马组织中的Notch1、Jagged1、Hes1及其mRNA表达。结果:造模后其他组较空白组,游泳不动时间增加(P<0.01)、旷场实验180 s内中间格停留时间及运动距离减少(P<0.01),而治疗28 d后电针组及西药组游泳不动时间减少(P<0.01)、旷场实验180 s内中间格停留时间及运动距离较模型组增加(P<0.01),西药组和电针组较模型组海马组织中MAP-2蛋白、Notch1蛋白及mRNA的表达量增加(P<0.05),Jagged1、Hes1蛋白及mRNA的表达量有所减少(P<0.05)。结论:以"肾脑相济"理论为指导的电针疗法可以通过调节海马Notch通路相关蛋白的表达,对围绝经期抑郁症起到治疗作用。 Objective: To observe the effect of electroacupuncture on expression of hippocampal Notch signaling pathway in rats with perimenopausal depression and explore the underlying mechanism. Methods: Forty-eight rats were divided into four groups:normal group,model group,western medicine group and electroacupuncture group. Establish a model of OVX + CUMS with rats. A sham operation group did not carry out the ovariectomy,and expect the normal group,other groups carried out the ovariectomy. The western medicine group was given clomipramine hydrochloride tablets. The electroacupuncture group was given acupuncture at Baihui( DU20),Shenshu( BL23) and( Sanyinjiao) SP6. The treatment was 28 days. Observe the result of behavioural changes,estrous cycle,body quality and the consumption of saccharose. Apply ELISA to test the content of MAP-2,ELISA and RT-PCR to test the content of Notch1,Jagged1,Hes1 and the mRNA. Results: After modeling,compared to the normal group,other groups' displacement distance and center stay time during 180 s reduced( P〈0. 01),and FST immobility time increased( P〈0. 01). However,after treatment,the electroacupuncture group and medicine group's displacement distance and center stay time and content of MAP-2,Notch1 and Notch1 mRNA in hippocampus increased( P〈0. 05),and contents of Jagged1,Hes1 and Jagged1 mRNA,Hes1 mRNA in hippocampus decreased( P〈0. 05). Conclusion: Guided by the theory of kidney and brain communication,electroacupuncture maybe accommodate the expression of hippocampal Notch signaling pathway in rats with perimenopausal depression to achieve the goal of treatment.
