摘要
为建立可供油桐尺蛾基础生物学研究的油桐尺蛾中肠上皮原代细胞系,采用组织块法、胰酶消化法和机械分散法分别对油桐尺蛾中肠上皮组织细胞进行体外培养。结果表明:组织块法培养效果最佳,细胞贴壁较好且生长旺盛。细胞移入培养瓶72 h可观察到细胞从组织块中迁移出来,呈细长梭形,随后数量增多形成网状集落,可维持良好的生长状态达60 d。说明油桐尺蛾中肠上皮原代细胞的最佳培养方法为组织块法。
This study was aim to establish midgut epithelial cellline of Buzura suppressaria Guenee for basic biology research. Midgut fragements of the moth were isolated and cultured with block explant culture technique, trypsin enzymatic digestion or mechanical dispersion in vitro. The results showed:the best cultivation was found from cells cultured with block explant culture technique, as- sessed by good cell adherence and growth. Epithelial cells started to migrate out from tissue blocks after 72 hours culture, shaped as fusiform and later reticular coloney growth with higher number. The cells could grow well until 60th day. We conclude that the explant culture technique using tissue blocks is best suited for primary cell culture of B. suppressaria Guenee midgut epithelial cells .
