载银载药纳米颗粒补片的抗菌效果研究 被引量:3

摘要 目的观察结合载银载药的补片在大鼠感染模型中的抗菌效果,对其抑制炎症反应的效果进行评价,进而探讨新型补片降低疝修补术后补片感染发生率的可行性。方法成年雄性SD大鼠50只,体重200~250 g,随机分为普通聚丙烯补片组(A组),新型载银载药纳米颗粒补片组(B组),并制备大鼠腹壁感染模型,将两种补片置于皮下,对比观察2组大鼠术后3、7、14、28 d的伤口感染及愈合情况,并于术后28 d处死大鼠,同时切取补片及其周围组织进行组织病理学观察。结果术后3 d观察到2组均出现炎症反应,伤口红肿、愈合欠佳,A组于术后7、14、28 d,均观察到伤口有脓液渗出,伤口愈合差;B组于术后7 d观察到与A组对比明显伤口无脓液渗出,愈合情况较好,并于14 d,达到完全愈合,无感染迹象。组织病理学观察,术后28 d,光镜下聚丙烯补片组依然有大量炎细胞浸润,可见毛细血管生长。结合载银载药纳米颗粒补片组组织可见少量炎细胞浸润。免疫组化IL-6分析,B组较A组炎症反应明显较轻。结论结合载银载药纳米颗粒补片在大鼠腹壁下感染模型中具有较好的抗菌效果,具有较好的临床应用前景,但其远期效果及生物安全性尚需进一步研究和评价。 Objective To observe the antibacterial effect of antibiotic- silver nanovehicles composite mesh in infection rat model, and to investigate the abilities of inhibition of inflammatory response, so as to investigate the feasibility of the application of the new type of mesh in reducing the hernia repair postoperative infection rate. Methods Fifty adult male SD rats ( weighting 200 to 250 g) were randomly divided into 2 groups ( n = 25 ) : ordinary polypropylene mesh group ( group A ) and new type of antibiotic-silver nanovehicles mesh group ( group B). The abdominal wall infection models were established in the rats, and place the two kinds of meshes on the fascia, respectively. Observe the situation of wound infection and healing at 3, 7, 14 and 28 days after operation. The animals were sacrificed at 28 days after operation for histological observations. Results Inflammation, wound swelling and poor healing were observed in both groups on day 3. In group A, pus exudation and poor wound healing were observed on day 7, day 14 and day 28. In group B, 7 days after operation, there was no significant wound infection in comparison with group A, the wound had no pus leaking and healed better. On the day 14, group A showed no signs of infection and healed well. The histological results showed that inflammatory cell infiltration, granulation tissue formation were observed in group A on day 28, compared with decreased inflammatory cell infiltration in group B. The level of IL-6 group B was smaller than group A. Conelusion Abdominal infection rat model with antibiotic-silver nanovehieles composite mesh, presents better wound healing and less inflammatory cell infiltration, compared with polypropylene mesh. The former has better clinical application prospects, but its long-term effect and biological safety need to be further studied and evaluated.
