健康人成功接种乙肝疫苗后的细胞免疫应答状态评价 被引量:2

Evaluation of the Cell Immunity Response of Healthy People Successfully Vaccinated with Hepatitis B Vaccine

摘要 目的:探究健康人成功接种乙肝疫苗后外周血单个核细胞(PBMCs)增殖及其Tbet、GATA-3、Foxp3、RORγt和Bcl-6五种CD4+T淋巴细胞亚群特异性转录因子的表达特征,评价细胞免疫应答状态。方法:本研究共采集64份外周血样本,分别来自43例疫苗免疫[HBs Ab(+)]和21例未疫苗免疫[HBs Ab(-)]的健康人。其中,有部分样本使用HBV重组核心抗原(r HBc Ag)进行诱导培养。通过CCK-8法检测细胞增殖情况;采用荧光定量PCR(FQ-PCR)法检测五种转录因子的m RNA表达水平。结果:HBs Ab(+)组五种转录因子基础表达水平均高于HBs Ab(-)组,其中GATA-3、Foxp3和R/F比值有显著差异(P<0.001)。经r HBc Ag诱导后,HBs Ab(-)组五种转录因子的m RNA表达水平较基础水平均有意义地增加(P<0.05);而在HBs Ab(+)组中,则均有不同程度降低,其中GATA-3显著降低(P=0.044);且诱导后HBs Ab(-)组五种转录因子表达水平均高于HBs Ab(+)组(P<0.05)。结论:乙肝疫苗免疫成功人群的细胞免疫在基础和体外再次应答水平上与非疫苗免疫健康者均存在明显的差别。 Objective: To observe the proliferation of peripheral blood mononuclear cells (PBMCs) and expressions of five transcription factors for CD4^+T lymphocyte subsets, including Tbet, GATA-3, Foxp3, RORγt and Bcl-6, and investigate the cell immunity response. Methods: In this study, blood samples were collected from 64 subjects, including 43 vaccine immunity subjects with HBsAb(+) and 21 non-vaccine immunity subjects with HBsAb(+). Among them, some PBMCs were induced by rHBcAg. By CCK-8, the proliferation of PBMC and rHBcAg-induced PBMC proliferation were detected. Using fluorescence quantitative PCR method, the basic expression levels of the five transcription factors and their rHBcAg-induced expression levels in PBMCs were detected. Results: The expressions of five transcription factors in HBsAb (+) group were higher than those in HBsAb (-) group, and GATA-3, Foxp3 and R/F were significantly different (P〈0.001). After rHBcAg induction, the expressions of five transcription factors in HBsAb (-) group were significantly increased (P〈0.05). In HBsAb (+) group, the expressions of five transcription factors were all decreased, of which GATA-3 was significantly decreased (P=0.044). After rHBcAg induction, five transcription factor expression of HBsAb (-) group were significantly higher than those of HBsAb(+) group(P〈0.05). Conelusion: HBV vaccinated population on the basis of immune cells, T cell responses were obviously different from non-vaccinated healthy controls.
