摘要
本研究利用特异性EST-SSR和SNP分子标记,对云南保山地区发现的荔枝新种质‘燎原’进行分子鉴定。通过研究其与现有367份荔枝种质(栽培品种与野生半野生种质)的亲缘关系,以明确其是否为一份新的种质,并揭示其亲本来源。基于34对EST-SSR标记和21对SNP标记的分析,燎原与其他367份荔枝种质的遗传相似系数介于0.289~0.849之间,遗传分化系数介于0.071~0.643之间,证实燎原应是一份与现有荔枝种质资源完全不同的新种质。进一步对燎原及其22份疑似亲本进行EST-SSR和SNP分析,并结合品种起源信息、果实成熟期以及植株表型特征,本研究推测‘燎原’是云南本地种质与外来广东栽培品种的杂交后代,即其母本为元矮,一个从本地半野生褐毛荔枝中选出的矮化变异品种,父本则为大造或中山状元红。
A novel litchi germplasm ‘Liaoyuan'found in Baoshan district of Yunnan province was identified by using specific EST-SSR and SNP molecular markers in this research. Through the analysis of genetic relationship between Liaoyuan and the existing 367 accessions of litchi germplasms(cultivated, wild and semi-wild germplasms),this study aimed to determine whether Liaoyuan was a novel germplasm, as well as to explore its candidate parent.Based on the analyses of 34 expressed sequence tag-simple sequence repeat(EST-SSR) markers and 21 single nucleotide polymorphism(SNP) markers, the genetic similarity coefficient between Liaoyuan and other germplasms was in the ranges of 0.289 to 0.849, and the genetic differentiation coefficient was in the ranges of 0.071 to 0.643.These results showed that Liaoyuan was a completely novel litchi germplasm, which was different from the known litchi resources. Through the further analysis of Liaoyuan and 22 suspected parents by using EST-SSR and SNP methods, combining with the information of germplasm origin, fructescence and phenotype characteristics, this research speculated Liaoyuan was a hybrid offspring of Yunnan local germplasm and exotic cultivar of Guangdong. The maternal parent of Liaoyuan might be Yuanai, a dwarf mutant accession selected from local semi-wild litchi, and its paternal parent might be Dazao or Zhongshanzhuanyuanhong.
