淮安地区流感病毒流行特征及分子进化研究 被引量:13

Epidemiological characterization and molecular evolution of the influenza virus prevalent in Huai'an

摘要 目的了解淮安地区流感病毒流行及变异特点。方法选择一个流感监测年度(2014-2015年)分离的流感病毒,进行HA1基因的扩增、测序,采用PHYLIP 3.6软件及popart1.7软件构建系统进化树及Network进化网络图,利用DnaSP5.1软件进行病毒多态性分析,利用MEGA 6.0软件进行氨基酸推导及相关特征分析。结果 2014-2015年淮安地区流行的流感病毒核苷酸同源性及多样度分别为98.2%~99.9%和0.009(新甲型H1N1),97.3%~99.9%和0.010(H3N2),98.7%~99.8%和0.009(B型Yamagata系),其中B型流感Yamgata系为淮安地区优势株。新甲型H1N1流感病毒淮安株在进化树上形成一个独立的进化亚枝,由A/Beijing-Xingcheng-Nan-Pian/SWL11619分化形成;H3N2流感病毒淮安株形成3个进化亚枝,由A/HK/7409/2014辐射进化形成;B型Yamgata系流感病毒淮安株形成2个进化亚枝,由B/Zhejiang-Liandou/11000/2014或B/Jiangsu-Hailing/11443/2014分化形成。与疫苗株相比,淮安地区各型流感病毒均发生的变异:K180Q、S220T(新甲型H1N1),N294K(H3N2),H137Q、N144K(B型Yamagata系)。结论淮安地区一个流行年度有两次流感病毒流行高峰,与南方流行趋势一致。 Objective To study the epidemiological characteristics and variation of the influenza virus. Methods HA1 gene sequences recovered from Huai'an isolates collected during the 2014-2015 influenza season were used to construct a phylogenetic tree and network diagram.Nucleotide polymorphisms of the influenza virus within a population were analyzed using DnaSP version 5.1.Amino acid sequences deduced from DNA were analyzed using MEGA5. Results During the 2014-2015 influenza season,the influenza virus present in Huai'an had a nucleotide similarity to H1N1(pdm09)of 98.2%-99.9% and a diversity of 0.009,a nucleotide similarity to H3N2 of 97.3-99.9% and a diversity of 0.010,and a nucleotide similarity to the influenza B Yamagata lineage of 98.7-99.8%and a diversity of 0.009.In addition,the influenza B virus Yamagata lineage was the predominant virus in Huai'an.Huai'an isolates of the influenza H1N1(pdm09)virus clustered together and formed a genetic lineage originating from A/Beijing-Xingcheng-Nan-Pian/SWL11619.Huai'an strains of the influenza H3N2 virus formed three lineages,with evolutionary branching from A/HK/7409/2014.Huai'an strains of the influenza B virus Yamagata lineage originated from B/Zhejiang-Liandou/11000/2014 or B/Jiangsu-Hailing/11443/2014.Compared to vaccine strains,Huai'an isolates of the influenza H1N1(pdm09)virus had K180 Qand S220T mutations,isolates of the influenza H3N2 virus had the N294 K mutation,and isolates of the influenza B virus Yamagata lineage had H137 Qand N144K mutations. Conclusion The prevalence of the influenza virus in Huai'an peaked twice,coinciding with trends in southern China from 2014-2015.
