摘要
目的观察扶正抑瘤汤含药血清对胶质瘤细胞共培养条件下骨髓间充质干细胞(BMSCs)的增殖、细胞周期及凋亡的影响,并探究其体外抗肿瘤作用。方法利用Transwell小室将BMSCs与胶质瘤细胞共培养,扶正抑瘤汤含药血清10%体积浓度干预。实验设4组:BMSCs对照组(BMSCs+空白对照血清)、BMSCs含药血清组(BMSCs+扶正抑瘤汤含药血清)、共培养对照组(BMSCs和胶质瘤细胞共培养+空白对照血清)、共培养含药血清组(BMSCs和胶质瘤细胞共培养+扶正抑瘤汤含药血清)。共培养72 h后,将BMSCs接种于96孔板中,分别培养24、48、72 h后,四甲基偶氮唑蓝(MTT)法检测BMSCs的细胞增殖情况,流式细胞仪检测BMSCs的细胞周期和凋亡率。结果共培养后BMSCs在胶质瘤微环境下增殖率不断上升(P<0.01),S期细胞数量增加(P<0.01),细胞凋亡率无明显变化;扶正抑瘤方含药血清干预后,BMSCs增殖受到显著抑制(P<0.01),S期细胞数量减少(P<0.01),细胞的凋亡率增加(P<0.01),且具有时间相关性。结论扶正抑瘤汤含药血清能抑制胶质瘤微环境下BMSCs的异常增殖,其机制可能与抑制DNA的合成和诱导细胞凋亡有关。
Objective To observe the effects of serum containing Fuzheng Yiliu Decoction on bone marrow mesenchymal stem cells (BMSCs)' proliferation, cell cycle and apoptosis when co-culturing with glioma, and explore its antitumor effect in vitro. Methods Co-culture of BMSCs and glioma cells was committed using Transwell. 10% volume concentration of serum containing Fuzheng Yiliu Decoction was employed to do the intervention experiment. There were 4 groups: BMSCs+blank control serum group, BMSCs+serum containing Fuzheng Yiliu Decoction group, co-cultured BMSCs cells and glioma cells + blank control serum group, co-cultured BMSCs cells and glioma cells + Fuzheng Yiliu Decoction containing serum group,. After incubated for 72 h, BMSCs were trypsinized and inoculated in 96 well plates, and then were cultured for 24 h, 48 h and 72 h, methyl thiazolyl tetrazolium method was used to assess the proliferation of BMSCs, the cell cycle and apoptosis rate of BMSCs were detected by flow cytometry. Results Compared with the BMSCs control group, the proliferation rate of BMSCs under the glioma microenvironment, was increased with time (P 〈 0.01), and also the proportion of cells in S phase was increased (P 〈 0.01),the apoptosis rate had no obvious change. Compared with the co-culture group, the proliferation rate of BMSCs in serum containing Fuzheng Yiliu Decoction group was decreased in a time-dependent manner (P 〈 0.01), the proportion of cells in S phase was reduced (P 〈 0.01), cell apoptosis rate was increased. Conclusion The serum containing Fuzheng Yiliu Decoction can obviously inhibit the proliferation of BMSCs, the molecular mechanisms may be related to inhibit DNA synthesis and induce cells apoptosis.
