川牛膝和怀牛膝多糖提取工艺研究 被引量:9

Research on the Extraction Process of Polysaccharides in Cyathulae Radix and Achyranthis Bidentatae Radix

摘要 以苯酚—硫酸法检测多糖含量,采用单因素考察、正交试验探究水煮提取法中料液比、提取时间、提取次数对川牛膝和怀牛膝多糖得率的影响并进行比较。结果表明,川牛膝多糖的最佳提取工艺为料液比1∶13(g/m L),提取时间2 h,提取次数3次;怀牛膝多糖的最佳提取工艺为料液比1∶13(g/m L),提取时间4 h,提取次数3次。两种多糖的最佳提取工艺条件不同,为后期的活性比较及入药选择研究奠定基础,为其工业化生产提供技术支持。 The different factors that affecting the yield of polysaccharides in Cyathulae Radix and Achyranthis Bidentatae Radix by water decocting extraction,including the ratio of solid to liquid,extraction time and extraction times,were detected and compared by the single factor investigation and the orthogonal experiment,while the contents of polysaccharides were detected by phenol-sulfuric acid method. The results showed that the best extraction process of the polysaccharide in Cyathulae Radix was as followes: the ratio of solid-liquid was 1 ∶ 13( g/m L),extracted for 2 h and repeated for 3 times,and the best extraction process of the polysaccharide in Achyranthis Bidentatae Radix was as followed: the ratio of solid-liquid was 1 ∶ 13( g/m L),extracted for 4 h and repeated for 3 times. The optimum conditions of the two kinds of polysaccharides are different,which will lay a foundation for the further studies such as active comparison and selection of medicine of the two polysaccharides,and the results could also provide technical support for industrialize production of the two polysaccharides.
