高血压大鼠线粒体DNA拷贝数与端粒长度的相关性研究 被引量:8

The relationship between leukocyte mitochondrial DNA copy number and telomere length in hypertensive rats model

摘要 目的探讨高血压大鼠外周血液白细胞线粒体DNA拷贝数和端粒长度的变化,以及二者之间的相关性。方法 SD大鼠40只,随机分为3组:高血压组(15只)、假手术组(15只)和空白对照组(10只)。高血压组采用双侧肾动脉后支结扎,术后予以8%NaCl饲料的方法构建高血压大鼠模型。选取术后4个月为观察终点,利用鼠尾血压计测量血压,随后行眶下静脉丛取血,提取血液基因组DNA,Real-time PCR法测量相对线粒体DNA拷贝数和端粒长度。结果术后4个月时与假手术组比较,高血压组大鼠血压显著升高[(178.36±10.21)比(128.47±8.74)mm Hg,P<0.01],线粒体DNA拷贝数显著增加[(1.49±0.43)比(1.09±0.51),P<0.05],端粒长度显著缩短[(0.83±0.23)比(1.04±0.29),P<0.05]。高血压组大鼠线粒体DNA拷贝数与端粒长度变化呈负相关(r=-0.589,P<0.05)。结论高血压大鼠模型可出现外周血白细胞线粒体DNA拷贝数升高和端粒长度缩短,且二者呈负相关。 Objective To explore the changes of peripheral leukocytes mitochondrial DNA copy number and telomere length in hypertensive rats, as well as the relationship between the two biomarkers. Methods 40 SD rats were randomly divided into 3 groups: hypertensive group(n=15), shame group(n=15) and control group(n=10). Hypertensive group was treated with ligations of posterior branches of bilateral renal arteries, and was fed with high salt diet(8%Na Cl). 4months after the operation, the blood pressure was measured by rat tail blood pressure gauge, genomic DNA was extracted from 1.5 m L blood which was acquired from inferior orbital venous plexus, mitochondrial DNA copy number and telomere length were measured by Real-time PCR. Results Compared with shame group at 4 months after the operation, the blood pressure was significantly increased [(178.36±10.21) vs(128.47±8.74) mm Hg,P〈0.01], the mitochondrial DNA copy number of hypertensive rats was significantly increased [(1.49±0.43) vs(1.09±0.51), P〈0.05], and the telomere length of hypertensive rats was significantly shortened [(0.83±0.23) vs(1.04±0.29), P〈0.05] in the hypertensive group. Mitochondrial DNA copy number was negatively related with telomere length(r=-0.589, P〈0.05) in the hypertensive group. Conclusion Increased mitochondrial DNA copy number and shortened telomere length are closely related with hypertensive in rats, and the two biomarkers appear a negative correlation.
