高效液相色谱法测定西柚不同组织中的10种柑橘黄酮类化合物 被引量:4

摘要 该文建立了圣草次苷、柚皮芸香苷、柚皮苷、橙皮苷、新橙皮苷、香风草苷、枸橘苷、甜橙黄酮、川皮苷和蜜橘黄酮等10种柑橘黄酮类化合物同时测定的高效液相色谱方法。色谱柱为Welch Ultimate XB-C18(5μm,4.6×150mm),采用甲醇-水(含0.1%甲酸)流动相体系,梯度洗脱,检测波长为280nm。方法学考察结果表明,对10种柑橘黄酮类化合物,提取效率均大于95%;在给定的范围内线性关系良好,相关系数r2均大于0.99;方法的精密度和重现性良好,相对标准偏差(RSD)分别小于5.4%和7.1%。该方法用于西柚不同组织中柑橘黄酮类化合物含量的测定,结果显示,柑橘黄酮类化合物在西柚果皮、囊衣和果瓣中的含量和分布差异显著。 A simple method was developed in this paper for simultaneous determination of 10 citrus flavonoids byhigh performance liquid chromatography,the compounds includes eriocitrin,narirutin,naringin,hesperidin,neohes-peridin,didymin,poncirin,sinensetin,nobiletion and tangeretin. The samples were separated by Welch UltimateXB-C18 column(5μm,4.6×150mm)using methanol and 0.1% formic acid aqueous solution as mobile phases by gra-dient elution detected by 280 nm. The results showed that there are good extraction efficiencies greater than 95%,good linear ranges with good correlation coefficients(r2)greater than 0.99,and good precisions and repeatabilitieswith relative standard deviations(RSD)less than 5.4% and 7.1% for all 10 citrus flavonoids. The method was usedfor citrus flavonoids determination in various parts of grapefruit,the results showed that the amounts and distribu-tions of citrus flavonoids have significant differences in grapefruit peel,utricle and pulp.
