摘要
目的观察右美托咪定的抗炎作用。方法在小鼠腋窝植入等质量的无菌棉球建立炎症模型,实验分对照组、地塞米松(dexamethasone(Dxm)5mg·kg-1)组、Test-L(右美托咪定(Dex)6.25μg·kg^(-1))、Test-M(Dex 12.5μg·kg^(-1))、Test-H(Dex 25μg·kg^(-1)),共5组,每组8只,植入当天开始腹腔注射给药,1日1次,共7d。ELISA测定小鼠血清TNF-α和IL-6的表达。结果右美托咪定能够抑制小鼠棉球肉芽组织的生成,能够抑制炎症因子TNF-α和IL-6的表达,且这种作用均呈现浓度依赖性。结论右美托咪定可能是通过抑制炎症因子的表达来发挥抑制棉球植入诱发的小鼠炎症反应。
OBJECTIVE To investigate the analgesic and anti-inflammatory effect of Dexmedetomidine. METHODS The effects of Dexmedetomidine on chronic inflammation was observed in the mouse inflammatory models of granuloma induced by cotton pellet. Mice were divided into five groups,Control group,dexamethasone group( Dxm 5mg·kg^(- 1)),the Test-L group( Dex 6. 25μg·kg^(-1)),the Test-M group( Dex 12. 5μg·kg^(-1)),and the Test-H group( Dex 25μg·kg^(-1)),eight mice in each group. The mice in granuloma model group induced by implantation of cotton pellet were administrated intraperitoneal injection for seven days,the levels of the inflammation factors TNF-α and IL-6 in mice serum were analyzed by ELISA. RESULTS Dexmedetomidine could significantly inhibit the formation of granulation tissue induced by cotton ball in mice. It could inhibit the expression of inflammation factors TNF-α and IL-6,and this effect was dependent in concentration. CONCLUSION Dexmedetomidine is probably by inhibiting the expression of inflammatory factor to inhibit the inflammatory response in mice induced by cotton ball implantation.
