摘要
目的:研究金桂外洗方对完全弗氏佐剂性炎症痛小鼠血清和局部前列腺素E2(PGE2)浓度的影响。方法:将体质量180~200g SPF级雌雄各20只KM小鼠在右后脚掌以完全弗氏佐剂法建立炎症模型,随机分为金桂外洗方外洗组(中药组)、扶他林溶液外洗组(西药组)、金桂外洗方+扶他林溶液外洗组(中西组)、造模组(阳性组)和空白组,1周后取小鼠血清和局部组织进行ELISA来测定PGE2.结果:1)血清PGE2浓度:阳性组〉西药组〉中药组〉中西组〉空白组,阳性组和西药组、中药组和中西组、中西组与空白组组间比较,组间差异有统计学意义(P〈0.01)。中药组、西药组、中西组、阳性组和空白组血清PGE2浓度分别是(1.02±0.20)pg/mL,(1.11±0.17)pg/mL,(0.85±0.15)pg/mL,(1.65±0.11)pg/mL,(0.56±0.14)pg/mL.2)局部组织PGE2浓度:阳性组〉西药组〉中药组〉中西组〉空白组。中西组与空白组组间比较,组间差异无统计学意义(P〉0.05);阳性组与西药组、西药组与和中药组、中药组和中西组组间比较,组间差异有统计学意义(P〈0.01)。中药组、西药组、中西组、阳性组和空白组局部组织PGE2浓度分别是(130.19±17.00)pg/mL,(210.29±14.50)pg/mL,(65.79±17.30)pg/mL,(788.00±95.15)pg/mL,(37.77±7.48)pg/mL.结论:金桂外洗方发挥消炎止痛的作用途径是通过降低血清和局部组织PGE2的浓度来实现,较扶他林作用明显。
Objective: To investigate the effect of Jingui Waixi decoction on PGE2 concentration in serum and local tissue in complete Freund's adjuvant (CFA) induced inflammatory pain mice model. Methods: CFA inflammation rats model was set up in the right rear foot of 180~200 g SPF level KM mice which were 20 male and 20 female respectively. Mice were randomly divided into five groups, which were J ingui Waixi decoction washed group (Chinese traditional medicine group, TCM group), voltaren washed group (modern medicine group), Jingui Waixi decoction added voltaren washed group (combination group), CFA model group (positive group) and saline treated group (blank group). The PGE2 concentra- tion of serum and local tissue was detected by ELISA kit one week after treatment. Results: 1)The serum PGE2 concen- tration were 1.02±0.20, 1. 11±0. 17, 0.85±0. 15, 1. 65±0. 11, and 0.56±0.14 (pg/mL) in the TCM group, modern medicine group, combination group, positive group, and blank group respectively. The concentration was followed by positive group〉modern medicine group〉TCM group〉 combination group〉blank group. The differences between posi tive group and modern medicine group, TCM group and combination group, combination group and blank group were sta- tistically significant (P〈0.01). 2)The local tissue PGE2 concentration were 130.19±17.00, 210.29±14.50, 65.79±17.30, 788.00±95. 15, and 37. 77±7.48 (pg/mL) in the TCM group, modern medicine group, combination group, positive group, and blank group respectively. The concentration was followed by positive group〉modern medicine group 〉TCM group〉 combination group〉 blank group. There was no significant difference between the combination group and the blank group (P〉0.05) ; The differences between positive group and modern medicine group, modern medicinegroup and TCM group, TCM group and combination group werre statistically significant (P〈 0. 01). Conclusion: The mechanism of Jingui Waixi decoction for inhibiting inflamma- tion and pain is though reducing PGE2 concentration both in serum and local tissue, and the effect is better than voltaren.
