菥蓂子乙醇提取物对H_2O_2诱导PC12细胞损伤的保护作用 被引量:4

摘要 目的:研究菥蓂子乙醇提取物对H2O2诱导损伤的PC12细胞保护作用及其作用机制。方法:用H2O2作用PC12细胞建立氧化应激损伤模型,采用MTT法测定细胞活力,活性氧检测试剂盒(DCFH-DA)检测细胞内活性氧,线粒体膜电位检测试剂盒(JC-1)检测细胞线粒体膜电位,TUNEL法检测细胞凋亡。结果:菥蓂子乙醇提取物能够保护H2O2诱导的PC12细胞活力损伤,抑制H2O2所致胞内ROS水平升高,抑制H2O2导致的线粒体膜电位下降,减少细胞凋亡的发生。结论:菥蓂子乙醇提取物能够抑制H2O2诱导PC12细胞氧化损伤,在神经细胞氧化损伤保护方面具有较好的应用价值。 Objective: To investigate the protective effect of the ethanol extracts of Thlaspi arvense L. seed on damage PC12 cells induced by H2O2 and its underlying mechanism. Methods: The cell viability was measured by MTT assay. The cell DNA damage was measured by TUNEL assay. The cell mitochondrial membrane potential was detected by the mitochondrial membrane potential detection kit( JC-1),and the cell reactive oxygen species( ROS) was detected by the reactive oxygen test kit.Results: The investigation showed that the ethanol extract of Thlaspi arvense L. seed can improve cell viability in damage PC12 cells induced by H2O2,inhibition of cellular DNA damage,stable mitochondrial membrane potential,inhibition of intracellular ROS increase. Conclusions: The results demonstrate that the Thlaspi arvense L. seed can suppress the oxidative damage in PC12 cells induced by H2O2 in multiple levels,which play a protective role in nerve cells.
