Expression of microRNA-616 in hepatocellular carcinoma and its clinical significance

摘要 目的:探讨micro RNA-616(mi R-616)在肝细胞癌(HCC)中的表达及临床意义。方法:应用实时定量PCR检测80例HCC患者的肿瘤组织与癌旁组织标本,以及正常肝细胞与不同分化程度的HCC细胞mi R-616表达;分析mi R-616表达与HCC患者临床病理因素及预后的关系;观察HCC细胞过表达或抑制mi R-616表达后侵袭及转移能力的变化。结果:与癌旁组织比较,HCC组织中mi R-616的表达明显升高,且复发患者明显高于非复发患者,有转移患者明显高于无转移患者(均P<0.05);所有HCC细胞株的mi R-616表达均明显高于正常肝细胞,且在高侵袭性HCC细胞株中的表达明显高于低侵袭性HCC细胞株(均P<0.05);mi R-616表达与HCC患者是否存在门静脉癌栓、Edmondson-Steiner分级、TNM分期有关(均P<0.05);mi R-616表达是影响HCC患者生存的独立危险因素(P<0.05),mi R-616高表达患者的总生存率和无病生存率均明显低于mi R-616低表达患者(均P<0.05)。HCC细胞过表达mi R-616后侵袭迁移能力明显增强,mi R-616表达抑制后侵袭迁移能力明显减弱(均P<0.05)。结论:mi R-616在HCC中表达升高,且mi R-616高表达与HCC不良预后密切相关。 Objective: To investigate the microRNA-616(miR-616) expression in hepatocellular carcinoma (HCC) and its clinical signiifcance. Methods: The miR-616 expressions in specimens of HCC and adjacent non-tumorous tissue from 80 HCC patients, as well as in normal hepatic cells and different HCC cell lines were determined by real-time PCR. hTe relations of miR-616 expression with pathologic factors and prognosis of HCC patients were analyzed and the changes&amp;nbsp;in invasion and metastatic abilities in HCC cells atfer miR-616 overexpression or inhibition were observed. Results: The miR-616 expression in HCC tissue was significantly elevated compared with adjacent non-tumorous liver tissue, and further, in recurrent cases was signiifcantly higher than that in non-recurrent cases and in cases with metastasis was signiifcantly higher than that in cases without metastasis(allP〈0.05). hTe miR-616 expressions were signiifcantly increased in all HCC cell lines compared with normal hepatic cells, which in highly invasive HCC cells were signiifcantly higher than those in lowly invasive HCC cells (allP〈0.05). hTe miR-616 expression was signiifcantly associated with the presence or absence of portal vein tumor thrombus, Edmondson-Steiner grade and TNM stage of HCC patients (allP〈0.05), and both overall survival and disease-free survival in HCC patients with high miR-616 expression were signiifcantly lower than those in patients with low miR-616 expression(bothP〈0.05). The invasive and metastatic abilities in HCC cells were significantly increased after miR-616 overexpression and were signiifcantly reduced atfer miR-616 inhibition (allP〈0.05). Conclusion: MiR-616 expression is increased in HCC, and the high miR-616 expression level is closely associated with poor prognosis.
