摘要 目的:探讨域含蛋白7A(THSD7A)基因在肝细胞癌(HCC)中的表达情况及临床意义。方法:分别用RT-PCR与免疫组化法检测30例新鲜HCC及癌旁组织标本中THSD7A m RNA表达及75例HCC及癌旁组织石蜡标本中THSD7A蛋白的表达,分析其与HCC患者临床病理因素及预后的关系。结果:与癌旁组织比较,在HCC组织中THSD7A m RNA表达下调(P<0.05);THSD7A在HCC组织中的阳性表达率明显低于癌旁组织(24.0%vs.87.0%,P<0.05)。THSD7A蛋白的表达与患者结节数目(P=0.011)、Edmondson-Steiner分级(P=0.013)、BCLC分期(P=0.048)有关。THSD7A蛋白低表达患者总体生存率(P=0.016)与无瘤生存率(P=0.013)均明显低于THSD7A高表达患者。结论:THSD7A在HCC中可能发挥抑癌基因的作用,THSD7A表达降低患者预后不良。 Objective: To investigate the domain containing 7A(THSD7A) expression in human hepatocellular carcinoma(HCC) and its clinical signii cance. Methods: The THSD7 A m RNA expression in 30 fresh specimens of HCC with paired and adjacent tissues,and protein expression in 75 parai n embedded specimens of HCC with paired adjacent tissues were detected by RT-PCR and immunohistochemical staining, respectively. h e relations of THSD7 A protein expression with clinicopathologic parameters and prognosis of HCC patients were analyzed.Results: The THSD7 A m RNA expression in HCC tissue was significantly down-regulated compared with adjacent tissue(P〈0.05), and the positive expression rate of THSD7 A protein in HCC tissue was signii cantly lower than that in adjacent tissue(24.0% vs. 87.0%, P〈0.05). h e THSD7 A protein expression was signii cantly related to the tumor numbers(P=0.011), Edmondson-Steiner grade(P=0.013) and BCLC stage(P=0.048) of the patients. Both overall survival and disease-free survival in patients with low THSD7 A expression were signii cantly lower than those in patients with high THSD7 A expression(P=0.016, P=0.013). Conclusion: THSD7 A may play a tumor-suppressor role in HCC, and those with decreased THSD7 A expressionhave a relatively poor prognosis.
