摘要
为探索红色诺卡氏菌细胞壁骨架中肽聚糖成分的抑瘤作用,通过超声波法碎红色诺卡氏菌细胞获得细胞壁,获得的细胞壁经过酶解、有机溶剂提取去除色素,得到细胞壁骨架(N-CWS),N-CWS再经去脂、酸解处理得到肽聚糖(N-CWS-PG)。对实验得到的提取物进行包括溶菌酶酶解处理、氨基酸分析、蛋白质含量的测定、脂类含量的测定、总糖含量的测定、氨基己糖的测定等化学鉴定。用实验提取的N-CWS-PG进行血清体外抑瘤试验,利用CCK-8法检测抑制效果;同时研究实验提取的N-CWS-PG对实体瘤小鼠的肿瘤抑制效果。实验结果表明;溶菌酶溶解实验和氨基酸分析结果证实所获取的目标物为肽聚糖,化学成分的测定结果表明其纯度达到97.20%;与对照组比较,血清学实验表明N-CWS-PG对S180细胞24,48 h的抑制率分别为9.72%,18.01%,实体瘤的抑制率为30.0%,实验结果证实N-CWS-PG具有抑制肿瘤细胞增殖的作用。
In order to explore the Nocardia rubra cell wall skeleton peptidoglycan inhibitory effect component,by ultrasonic broken Nocardia rubra cells obtaining the cell wall; the cell wall obtained through enzymatic hydrolysis and the organic solvent extraction to remove the pigment,obtaining the cell wall skeleton( N-CWS). Then the N-CWS by degreasing,acid hydrolysis process to obtain the Nocardia rubra cell wall skeleton peptidoglycan( N-CWS-PG). The extract was subjected to experiments including lysozyme enzymatic treatment,amino acid analysis,chemical identification determination of protein content,lipid content determination,determination of total sugar content,hexosamine determination and so on. The experiments were carried out by extracting N-CWS-PG serum tumor growth in vitro test to detect the use of inhibitory effect of CCK-8 method,and research experiments extracted N-CWS-PG tumor solid tumor inhibitory effect. The experimental results show: lysozyme dissolved target experiments and amino acid analysis confirmed the acquired as peptidoglycan. The measurement results showed that the chemical components was of the purity of97. 20%; compared with the control group,the experimental results show that serum N-CWS-PG on S180 cells 24 h,48 h inhibition rates were 9. 72% and 18. 01%; the solid tumor inhibitory rate was 30. 0%. The experimental results confirmed N-CWS-PG inhibiting tumor cell proliferation.
