摘要
目的探讨肽基脯氨酰顺反异构酶B(PPIB)对大鼠胰岛β细胞中胰岛素合成的影响。方法用不同浓度环孢素A 0、0.1、0.5、1、2.5、5、7.5、10、12.5和15μmol/L处理INS1细胞,采用MTT法检测INS1细胞活力,Western blot法检测INS1细胞中PPIB和胰岛素表达水平。结果环孢素A呈时间和剂量依赖性地抑制INS1细胞活力,降低PPIB和胰岛素含量(P<0.05或P<0.01)。结论环孢素A可能通过下调PPIB表达抑制胰岛细胞中的胰岛素合成。
Objective To investigate the effect of peptidly-prolyl isomerase B(PPIB)on the synthesis of insulin in rat isletβcells.Methods The INS1 cells were treated with different concentrations of cyclosporin A 0,0.1,0.5,1,2.5,5,7.5,10,12.5 and 15 μmol/L.The cell viability and insulin expression were detected by MTT assay and Western blot,respectively.Results Cyclosporin A inhibited cell viability in dose-and time-dependent manners and decreased the expressions of PPIB and insulin(P〈0.05 or P〈0.01).Conclusion Cyclosporin A can reduce insulin production in rat islet β cells probably by down-regulating PPIB expression.
