摘要
目的建立高效液相非接触电导检测法测定盐酸二甲双胍片中盐酸二甲双胍含量的新方法 。方法 色谱系统采用Ucleosil C18柱(4.6×250 mm);流动相:磷酸二氢铵溶液(称取磷酸二氢铵17 g,加水1000 ml溶解,用磷酸调节p H至3.0);流速:1.0 ml/min;进样量:10μl;柱温:35℃。用非接触电导检测器对盐酸二甲双胍进行检测。结果 盐酸二甲双胍进样量在0.10-1.04μg范围内与峰面积响应值呈良好线性关系(r=0.9960),平均回收率(n=9)为101.0%;RSD为0.26%(n=6)。结论 该方法简便、灵敏、重复性好,适用于盐酸二甲双胍的含量测定,结果满意。
Objective To establish a new method for the rapid determination of mefformin hydrochloride in metformin hydrochloride tablets by HPLC determination with contaetless conductivity detection. Methods HPLC method was carried on a colmn of Ucleosil Cls (4.6x250 mm)at column temperature of 35℃ ,with the mobile phase of ammonium dihydrogen phosphate solution (ammonium dihydrogen phosphate solution 17 g with 1000 ml water,adjusting pH to 3.5 with phosphoric acid),at flow rate of 1.0 ml/min,detection of metformin hydrochloride by contactless conductivity detection. Results The linear range of metformin hydrochloride was 0.10-1.04 μg (r=0.9960) and the average recovery rate was 101.9% with RSD 0.26% (n=6). Conclusion The method is convenient,rapid and reproducible,and it can be used for rapid determination of metformin hydrochloride with satisfactory result.
