摘要
目的建立高效毛细管电泳法测定注射用头孢噻肟钠含量的方法.方法采用涂层石英毛细管(60 cm×75 μm,有效长度53 cm);运行缓冲液为30 mmol/L硼砂溶液(pH 9.2),高压进样5 s,分离电压12 kV,柱温25℃,检测波长为254 nm,地塞米松磷酸钠为内标.结果头孢噻肟钠在6~30 μg/ml浓度范围内线性关系良好(r=0.999 7).结论此方法简单、快捷、灵敏,可用于注射用头孢噻肟钠的含量测定.
Purpose To determine the content of cefotaxime sodium for injection. Methods the content of cefotaxime sodium for injection was determined at 254 nm using high performance capillary electrophoresis (HPCE), under the best conditions achieved with a fused-silica capillary tube (60 cm × 75 /μm , effective length of 53 cm) and 30 mmol/L borax (pH 9.2) at a constant voltage of 12 kV and a sample time of 5 s at 25 ℃. Dexamethasone sodium phosphate was internal standard. Results The calibration curve displayed good linear realationship within cefotaxime sodium concentration range of 6 to 30μg/ml (r = 0. 9997). The average recovery of cefotaxime sodium was 98.93 % ( n = 5, RSD = 1 . 98 % ). Conclusion HPCE is simple, quick and sensitive in determination of cefotaxime sodium for injection.
