摘要
探讨提取白术根茎总RNA的最优方案。以新鲜白术的根茎为实验材料,采用三种方法提取其根茎总RNA。用紫外分光光度法和琼脂糖凝胶电泳检测RNA样品完整性,用RT-PCR方法验证RNA质量。用多糖多酚植物总RNA提取试剂盒的方法,获得了完整性最好、质量最高的总RNA样品。提取白术根茎总RNA的三种方法中,多糖多酚植物总RNA提取试剂盒的方法最理想。
Objective： to investigate the best way to extract total RNA from fresh rhizome of Atractylodes macrocephala.Method： taken fresh rhizome of A. macrocephal as material, the total RNA was extracted with three methods. We detected the integrity of total RNA samples, using ultraviolet spectrophotometry and agarose gel electrophoresis, and RT-PCR method for testing the quality of total RNA samples. Result： the analysis of agarose gel electrophoresis and RT-PCR amplification showed that,RNA sample of the highest quality and the best integrity was obtained by the method of polysaccharide polyphenol plant total RNA extraction kit. Conclusion： the method of polysaccharide and polyphenols plant total RNA extraction kit was the best among the three kinds of extraction methods of RNA from fresh rhizome of A. macrocephala.
