正交试验优选丁香叶中总酚酸制备工艺

Optimization of Total Phenolic Acid in Clove Leaves by Orthogonal Test

摘要 目的:优选丁香叶中总酚酸最佳水提取乙醇沉降工艺。方法:以出膏率、原儿茶酸、咖啡酸的量为评价指标,选取加水倍量、煎煮时间和煎煮次数为考察因素,采用正交试验优选丁香叶中总酚酸的最佳水提工艺;选取醇沉浓度、醇沉时间、醇沉温度为考察因素,采用正交试验优选丁香叶提取液的乙醇沉降工艺。结果:优选的最佳提取工艺为加10倍量的水,煎煮3次,每次1 h,最佳醇沉工艺为含醇量为70%,温度60℃,静置12 h。结论:该工艺简单稳定,可作为丁香叶工业化的提取工艺。 Objective:Preferred lilac leaves total phenolic acids optimum water extracts by ethanol precipitation technology. Methods:Rate of paste,the protocatechuate,the caffeic acid amount of the evaluation index,selected fold amount water,decoction time and decocting times for the study factors,using by orthogonal test lilac leaves of total phenolic acid the best water extraction process;selected concentration of alcohol precipitation,alcohol precipitation time,alcohol precipitation temperature was study factors,the use of orthogonal test lilac leaves extract by ethanol precipitation technology. Results:Preferred optimum extraction technology of plus 10 times the amount of waterdecocted and decocting 3 times,each time 1 hour,best alcohol precipitation process was alcohol content of 70%,temperature 60 ℃,allowed to stand 12 h. Conclusions:The technology is simple and stable,can be used as the extraction process of clove leaf industrialization.
