摘要
本实验利用Gateway技术构建了Ubiquitin启动子驱动的高羊茅Fa SGR基因的RNAi载体p ANDA-B-870,该载体具有潮霉素抗性,适合于单子叶植物遗传转化。通过电击法将p ANDA-B-870载体转入到根癌农杆菌EHA105感受态细胞中,验证转化成功。Fa SGR基因RNAi载体的构建为进一步探讨Fa SGR基因功能及SGR滞绿基因在高羊茅遗传改良中的应用提供了技术支持。
In this study,RNAi vector p ANDA-B-870 with hygromycin resistance for tall fescue Fa SGR gene drove by Ubiquitin promoter was constructed using the Gateway technology,which was suitable for genetic transformation of monocots. The p ANDA-B-870 vector was transformed into Agrobacterium tumefaciens EHA105 competent cells by electroporation method,the transformation was confirmed by PCR. The construction of RNAi vector for Fa SGR gene provided technical support for further study of Fa SGR gene's function and the application of SGR of tall fescue in genetic improvement.
