胡椒酸己二胺对高脂血症大鼠降低血脂的实验研究

摘要 目的研究胡椒酸己二胺(PAH)对实验性高脂血症大鼠的降血脂作用。方法雄性SD大鼠78只,适应性饲养1w后,测定基础血清胆固醇(TC)和甘油三酯(TG),随机抽取12只作为正常对照组,给予普通饲料,余作为造模组喂以高脂饲料。喂养4w后,再次检测血清TC和TG,比较造模前、后的血脂水平,以TC、TG显著升高为造模成功。将造模成功的大鼠按血清TC和TG水平分层,并随机分为高脂模型组、辛伐他汀阳性药物组(10mg/kg体质量)和PAH高剂量(10mg/kg体质量)、中剂量(5mg/kg体质量)、低剂量(2.5mg/kg体质量)组。每日灌胃给予各受试物,第4周末,麻醉动物,采集血清,测定TC、TG、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C),同时制作石蜡切片,光镜观察各组大鼠肝脏组织形态变化。结果 PAH中、高剂量组血清TC水平较高脂模型组低,LDL-C水平较阳性药物组降低,差异均有统计学意义(P<0.05);PAH各剂量组血清TG、LDL-C水平均较高脂模型组低,差异具有统计学意义(P<0.05);低剂量组HDL-C水平较高,差异具有统计学意义(P<0.05)。PAH各剂量组血清TC、LDL-C水平存在剂量-效应关系,在高剂量时差异最为显著。中剂量PAH可明显改善肝细胞肿胀情况,对肝细胞有一定的保护作用。结论胡椒酸己二胺具有降血脂以及保护肝脏的作用。 Objective To study the effect of pepper acid hexamethylenediamine(PAH)on experimental hyperlipidemia rats in reducing blood lipids.Methods 78 male SD rats,determined basal serum cholesterol(TC)and triglyceride(TG)level,randomly selected 12 as control group received regular diet,and others as model group fed with high-fat diet for 4 weeks.According to the serum TC and TG levels detecting,the successful models were stratified and randomized into high fat model group,simvastatin group(10mg/kg body weight)and PAH high dose(10mg/kg body weight),middle dose(5mg/kg body weight),and low dose(2.5mg/kg body weight)groups,then with drugs oral delivery for 4 weeks.Finally,collect blood samples and test TC,TG,HDL-C,and LDL-C,and paraffin sections were made to observe the morphological changes of the liver tissues of rats in all groups by light microscope.Results PAH middle dose and high dose group,the serum TC level were lower than the hyperlipidemia model group,LDL-C levels were reduced compared with simvastatin group with statistical significance(P〈0.05);PAH 3 dose groups,serum TG,LDL-C levels were lower than the model group with statistical significance(P〈0.05),while HDL-C level of low dose group was higher with statistical significance(P〈0.05).There was a dose effect relationship between TC and LDL-C levels in serum of the 3 dose groups of PAH,and the difference was the most significant at high dose.The middle dose group significantly improved liver cell edema,so existed certain protective effect on liver cells.Conclusion PAH had a conspicuous effect on antiatheroscloresis and protection of liver.
