摘要 目的观察AKT-GSK-3β信号转导通路对化疗后肝癌细胞凋亡的影响,进一步探讨该通路在肝癌细胞化疗敏感性中的调控作用。方法应用细胞培养与Western blot技术,检测肝癌细胞化疗后凋亡情况并分析其与AKT-GSK-3β信号通路活化的关系,通过特异性阻断信号通路方法,对AKT-GSK-3β通路调节肝癌细胞化疗敏感性进行验证。结果肝癌细胞经奥沙利铂化疗后发生凋亡,表现为BAX蛋白表达增加和Bcl-2蛋白表达减少,同时AKT-GSK-3β通路磷酸化激活。阻断该通路后,化疗后肝癌细胞凋亡明显增加。结论 AKT-GSK-3β信号途径可以下调肝癌细胞化疗敏感性,与其化疗效果有密切联系。 Objective To investigate the effects of AKT-GSK-3β signaling pathway on hepatocellular carcinoma cell apoptosis after chemotherapy. Methods Cell culture and western blot were used to detect apoptosis of hepatocellular carcinoma cells induced by oxaliplatin,and the relationship between apoptosis and AKT-GSK-3βsignaling pathway activation were analyzed. Through inhibiting signaling pathway specifically,regulation effects of AKT-GSK-3β signaling pathway on sensitivity of hepatocellular carcinoma cells to chemotherapy was verified. Results Hepatocellular carcinoma cells appeared apoptosis after chemotherapy,showing increase of BAX protein and decrease of Bcl-2 protein as well as phosphorylation of AKT-GSK-3β signaling pathway.However,Hepatocellular carcinoma cells apoptosis increased obviously after chemotherapy. Conclusion AKTGSK-3β signaling pathway could down-regulate sensitivity of hepatocellular carcinoma cells to chemotherapy and may have closely relationship with therapeutic effect.
