Effect of recombinant human growth hormone on burn wound healing in rat model

摘要 [目的]观察重组人生长激素(rhGH)对大鼠烧伤创面模型伤口愈合的影响.[方法]选择30只大鼠建立皮肤烧伤模型后,随机分为正常对照组、rhGH大、小剂量组.每日定时腹部皮下注射给予rhGH大、小剂量组rhGH 0.30,0.15U/mL至创面湿润,每日滴加给予正常对照组生理盐水2mL/kg至创面湿润,治疗14d后观察大鼠一般状态及烧伤创面愈合情况,并监测药物不良反应发生情况.[结果]rhGH大、小剂量组创面愈合时间分别为(19.3±4.5)d,(18.9±4.2)d,与正常对照组(28.9±5.8)d比较明显缩短(P<0.01).用药后第1,3,5周的观察结果显示,rhGH大、小剂量组血糖值均在正常范围内,未出现明显神志改变、多饮多食、活动量减少、体质量猛增等不良反应.[结论]rhGH可有效促进大鼠烧伤创面的愈合,且用药期间均未发生不良反应. OBJECTIVE To study the effect of recombinant human growth hormone(rhGH)on burn wound healing in rat model.METHODSSkin burn models in 30 rats were established,and were randomly divided into groups of normal control,large and small dosage of rhGH.0.30 and 0.15U/mL of rhGH in large and small dosage groups of rhGH were daily administrated by abdomen subcutaneous injection on time to wound surface moist,and 2mL/kg of normal saline in normal control group were daily bedewed to wound surface to moist for 14 days.General state of rats and burn wound healing was observed,and the occurrence of adverse drug reactions was monitored.RESULTS(19.3±4.5)days and(18.9±4.20 days of wound healing time in large and small dosage groups of rhGH shortened significantly than(28.9±5.8)days in normal control group(P〈0.01).After treatment for 1,3and 5weeks,mean value of blood glucose in large and small dosage groups of rhGH was within normal range,and the adverse drug reactions,significantly altered mental status,polydipsia and polyphagia,reducing activity levels and increase of body weight et al,were not observed.CONCLUSIONRhGH can effectively promote the healing of burn wound in rats,and does not induce the adverse drug reactions.
