牛蒡苷对弓形虫热休克蛋白70诱导弓形虫感染小鼠急性肝损伤的保护作用 被引量:3

Protective effects of the arctiin on Toxoplasma gondii-derived heat shock protein 70-induced acute liver injury in T.gondii-infected mice

摘要 [目的]探讨牛蒡苷(ARC)对弓形虫热休克蛋白70(T.g.HSP70)诱导弓形虫感染小鼠急性肝损伤的保护作用及其机制.[方法]将KM雌性小鼠随机分为正常组、模型组、ARC组、磺胺嘧啶钠阳性对照(PC)组.于弓形虫感染后第4d起给予药物治疗,连续5d.感染后第7d腹腔注射给予T.g.HSP70,次日末次给药4h后采集颈动脉血处死小鼠,取肝称质量,计算肝脏指数;采用比色法测定谷丙转氨酶(ALT)、谷草转氨酶(AST)水平、丙二醛(MDA)含量及超氧化物歧化酶(SOD)活性;ELISA法测定细胞白细胞介素-1β(IL-1β)、肿瘤坏死因子α(TNF-α)水平;采用逆转录-聚合酶链式反应(RT-PCR)测定血小板活化因子(PAF-AH)mRNA的表达水平.[结果]与正常组比较,模型组ALT,AST水平和肝脏指数明显升高(P<0.05,P<0.01).与模型组比较,ARC组和PC组ALT,AST水平和MDA,IL-1β,TNF-α含量及PAF-AH mRNA表达水平均明显降低(P<0.05,P<0.01),SOD活性明显升高(P<0.05).ARC组与PC组上述各项指标间差异均无统计学意义(P>0.05).[结论]ARC对T.g.HSP70诱导弓形虫感染小鼠急性肝损伤具有保护作用,其机制可能与抗脂质过氧化及抑制炎症介质的产生有关. OBJECTIVE To study the protective effects and its mechanism of arctiin(ARC)on Toxoplasma gondii-derived heat shock protein 70(T.g.HSP70)-induced acute liver injury in T.gondii-infected mice.METHODS KM female mice were randomly divided into groups of normal,model,ARC and PC.Post infection(PI)of T.gondii at 4th day,the experimental groups were treated with medicine,and continuous infection for 5 days at 7th day,the mice were administrated by intraperitoneal injection with T.g.HSP70,and after end of administration medicine for 4h at the next day,the mice were executed by collecting arterial blood,and liver was taken out to weigh up its weight and to calculate the liver index.The contents of AST,ALT,MDA and activity of SOD were detected by colorimetry,of IL-1βand TNF-αELISA,and the expression of PAF-AH mRNA by RT-PCR.RESULTS The contents of ALT and AST and index of liver were obviously increased in the model group than in the normal group(P〈0.05,P〈0.01),and the index of liver,the contents of ALT,AST,MDA,IL-1β,TNF-α,and the expression of PAF-AH mRNA in the ARC and PC groups were significantly decreased than that in the model group(P〈0.05,P〈0.01),and the activity of SOD was increased in both ARC and PC groups(P〈0.01),and above indexes had no statistical significance between ARC and PC(P〈0.05).CONCLUSION ARC has a protective effect on T.g.HSP70-induced acute liver injury in T.gondii-infected mice,and its protective mechanisms maybe related to anti-lipid peroxidation and inhibition of inflammatory mediators.
