子宫内膜癌宫腔诊刮的抑癌基因病理学表达及其影响因素分析 被引量:5

摘要 目的分析子宫内膜癌宫腔诊刮的抑癌基因病理学表达及其影响因素。方法将接受子宫内膜宫腔诊刮治疗的150例纳入研究对象,85例子宫内膜癌患者作为观察组,65例正常无病变者作为对照组。比较两组受试者抑癌基因p16、PTEN、BRCA-1的mRNA含量和蛋白含量,并分析病理分期和浸润深度对抑癌基因表达的影响。结果观察组患者子宫内膜组织中p16基因、PTEN基因、BRCA-1基因的mRNA含量和蛋白含量均低于对照组(32.85±5.37vs.100±14.85,29.62±3.85 vs.100±16.27,24.51±3.26 vs.100±15.42;29.94±4.28 vs.100±14.19,31.47±4.85vs.100±15.28,25.92±3.08 vs.100±16.41);不同病理分期、浸润深度的患者,抑癌基因表达情况有差异,病理分期越高、浸润程度越深,p16基因、PTEN基因、BRCA-1基因的mRNA含量和蛋白含量越低。结论子宫内膜癌组织中抑癌基因p16、PTEN、BRCA-1表达量显著降低,且表达情况受到病理分期、浸润深度的影响。 Objective To analyze the pathological expression of tumor suppression gene in intrauterine diagnostic curettage for endometrial cancer and its influencing factors. Methods A total of 85 patients with endometrial cancer performed intrauterine diagnostic curettage were enrolled in observation group and 65 normal women were enrolled in control group. Then mRNA and protein contents of tumor suppressor gene p16,PTEN and BRCA- 1 were compared and the influence of pathological stage and depth of invasion on expression of tumor suppressor gene had been analyzed. Results The content of mRNA and protein content of p16 gene,PTEN gene,BRCA- 1 gene in uterine endometrial tissue of patients in observation group were significantly lower than those of control group( 32. 85 ± 5. 37 vs. 100 ± 14. 85,29. 62 ± 3. 85 vs. 100 ± 16. 27,24. 51± 3. 26 vs. 100 ± 15. 42; 29. 94 ± 4. 28 vs. 100 ± 14. 19,31. 47 ± 4. 85 vs. 100 ± 15. 28,25. 92 ± 3. 08 vs. 100 ± 16. 41). The expression level of tumor suppressor gene in patients with different pathological stages and depth of invasion was different. As the increase in pathological stage and depth of invasion,the contents of mRNA and protein of p16 gene,PTEN gene and BRCA- 1 gene would be lower. Conclusion The expression levels of tumor suppressor gene p16,PTEN and BRCA- 1 are significantly decreased in uterine endometrial carcinoma and their expression levels are influenced by pathological stage and depth of invasion of the tumor.
