摘要
目的探讨胃腺癌病人血浆中miR-199a-5p、miR-200c-3p的表达及其诊断和预后判断的价值。方法选取2014年5月至2015年4月胃腺癌病人50例为观察组,同期入选体检健康者50例为研究对照,应用实时荧光定量PCR技术检测两组研究对照血浆中的miR-199a-5p和miR-200c-3p的表达水平情况;比较不同分化程度、淋巴结转移情况的胃腺癌病人的血浆中miR-199a-5p、miR-200c-3p表达水平。结果观察组的miR-199a-5p表达水平显著低于对照组,差异有统计学意义(P<0.05);观察组的miR-200c-3p表达水平显著高于对照组,差异有统计学意义(P<0.05);不同分化程度、淋巴结转移情况的胃腺癌病人的血浆miR-199a-5p、miR-200c-3p表达水平差异有统计学意义(P<0.05)。结论胃腺癌病人血浆miR-199a-5p、miR-200c-3p表达水平与肿瘤分化程度、淋巴结转移情况密切相关,可用于评估胃腺癌的病情严重程度及预后。
Objective To explore the diagnostic and prognostic significance of expression of plasma miR-199a-5p and miR-200c-3p in patients with gastric cancer. Methods A total of 50 patients with gastric cancer in this hospital during May 2014 to April 2015 were allocated in observation group,and 50 healthy persons in the same period were selected as control group,the expression levels of plasma miR-200c-3p and miR-199a-5p in these two groups were detected by real-time fluorescent quantitative PCR; and the expression levels of plasma miR-200c-3p and miR-199a-5p in patients with gastric cancer at different degree of differentiation and metastasis in lymph nodes. Results The expression level of miR-199a-5p in patients of observation group was significantly lower than that of controls in control group,the difference was statistically significant（ P〈0. 05）; and the expression level of miR-200c-3p in patients of observation group was significantly higher than that of controls（ P〈0. 05）,and the expression levels of miR-199a-5p and miR-200c-3p in gastric adenocarcinoma patients with different degree of differentiation and metastasis in lymph nodes were significantly different（ P〈0. 05）. Conclusion The expression levels of miR-200c-3p and miR-199a-5p in plasma of patients with gastric cancer were detected,and they are closely related with the degree of differentiation of cancer and metastasis in lymph nodes,and their detection can be applied for evaluation of the severity and prognosis of patients with gastric cancer.
