Simultaneous determination of eight sexual hormones in cosmetics by high performance liquid chromatography with diode array and fluorescence detectors

摘要 建立了高效液相色谱(HPLC)-二极管阵列(DAD)/荧光检测器(FLD)串联测定化妆品中雌三醇、雌二醇、睾酮、炔雌醇、己烯雌酚、乙酸甲地孕酮、孕酮和丙酸睾酮8种性激素的分析方法。所有形态的化妆品样品均以甲醇超声提取进行前处理,乳膏状样品再经HLB固相萃取柱富集净化,以乙腈/水为流动相梯度洗脱,经ZORBAX Eclipse XDB-C18柱分离,DAD-FLD串联法检测。8种性激素获得了满意的分离并排除了样品基质的干扰。方法检出限为0.1-0.5μg/g,平均加标回收率为90.3%-102.9%(n=6),相对标准偏差为0.4%-4.8%(n=6)。方法可用于化妆品中雌三醇等8种痕量性激素的同时测定。 A method of simultaneous determination of 8 sexual hormones ( estriol, estradiol, testosterone, 17α-ethinylestradiol, diethylstilbestrol, megestrol-17-acetate, progesterone and testosterone propionate) in cosmetics has been developed by high performance liquid chromatography with diode array and fluorescence detections. All of the cosmetic samples were extracted with methanol by ultrasonic assisted extraction and the extracts of the cream samples were purified by HLB solid phase extraction column. The extracted supernatants were satisfactorily separated in an Agilent ZORBAX XDB-CI8 column using acetonitrile-water as mobile phase for gradient elution. The simultaneous measurements for the substances were completed by a diode array detector (DAD) and a fluorescence detector (FLD), and external standard method was adopted for quantification. The limits of detection for the method were 0. 1 -0. 5 μg/g. The average recoveries were 90. 3% - 102. 9% and the relative standard deviations (RSD) were between 0. 4% and 4. 8% (n = 6). The method is simple and sensitive, and can be used for the quantitative detection of trace amounts of the sexual hormones in cosmetics.
