摘要
目的研究"葛根总黄酮-黄连生物碱"3:8配伍对链脲佐菌素(STZ)复合高脂饲料致2型糖尿病大鼠模型的药效学作用,为"葛根总黄酮-黄连生物碱"组分配伍在防治2型糖尿病创新药物研究与开发提供一定的科学依据.方法建立链脲佐菌素(STZ)复合高脂饲料诱导大鼠2型糖尿病动物模型,分别测定大鼠的空腹血糖(FBG)、血清胰岛素(Ins)以及血清胆固醇(TC)、甘油三酯(TG)的含量,并观察药物对大鼠胰腺组织病理的影响.结果葛根黄连有效组分配伍后,能明显降低模型大鼠的FBG,增加Ins的含量,有效增加模型大鼠机体胰岛素的敏感性,改善胰岛素抵抗;同时,药物能明显降低模型动物血清TC和TG的含量;胰腺组织病理HE染色观察显示,葛根黄连有效组分在一定程度上改善大鼠胰腺组织的损伤,增加胰腺中胰岛的数量,改善腺泡及胰岛的萎缩程度.结论葛根黄连有效组分配伍具有确切调节糖代谢、脂质代谢的作用,改善胰岛素抵抗,有效保护胰岛β细胞.
Objective To study the pharmacodynarnics effect of radix Puerariae and Rhizomacoptidis 3 : 8 on type 2 diabetes rat model, and to provide scientific data for the development of innovative drugs for the type 2 diabetes. Methods Using rat model of type 2 diabetes induced by STZ and high fat diet to determine FBG,serum insulin(Ins) ,serum cholesterol(TC) and triglyceride (TG), and to study the effect of drug on the pathological changes of rats. Results The active components of Puerarin and Rhizo- maeoptidis significantly reduced the FBG, increased the content of Ins, enhanced the sensitivity of insulin in the rats, improved insulin resistance, and significantly reduced serum cholesterol (TC) and triglyeeride (TG) in diabetic animal models. It also had a certain effect on the repair of pancreatic tissue, increased the number of pancreatic islets, and improved the pancreatic foam and pancreatic islet atrophy. Conclusion The active component compatibility of Puerarin and Rhizomacoptidis regulates the glu- cose and lipid metabolism, and improves insulin resistance and protects β - cells of pancreatic islet effectively.
