摘要
本研究结果表明,米托蒽醌对小鼠的LD_(50)与阿霉素相近,对外周血降白细胞作用强于阿霉素,心肌标本电镜观察显示损伤较阿霉素轻。对犬大剂量静脉推注可使其血压下降、心率减慢。
Acute toxicity of mitoxantrone (DHAQ)synthetized by Department of Organic Che-mistry, School of Pharmacy, WCUMS wasstudied in mice and dogs. The toxic effectswere compared with those of adrinamycin(ADR). The LD_(50) were 6.6±1.3 and 7.1±1.6 mg/kg DHAQ, 7.9 ±2.1 and 10.7± 2.2mg/kg ADR (P= 0.95) respectively on ob-servation with a single dose i.v. and i.p.,for 21 days in Kunming mice. The toxicityappeared to be delayed with DHAQ andADR. The earliest death occurred on 4th dayafter treatment in mice. In the third weekstill a few died. There were obvious decreases in WBCcounts in the peripheral blood of micereceiving either 1-2.5mg/kg DHAQ or 2.5-5mg/kg ADR with a single dose i.p.. TheWBC count could return to normality after3 weeks with the withdrawal of the drugs. On electron microscopic examination ofmyocardial samples from mice 4 days aftera single dose of 20 mg/kg DHAQ i.P., mi-tochondria swelling and vacuoles formationin some cells were shown. Besides theabove-mentiond changes, some myocytesexhibited disorientation and loss of myofi-laments in the mice which received equi-valent dose of ADR. In dogs anaesthetized a temporary fal-ling of blood pressure and slowing of heartrate were observed following 5-10 mg/kgDHAQ i. p.. When a dose of 50-100mg/kgwas given, the blood pressure dropped con-tinuously until death.
