复合蛋白酶和风味蛋白酶双酶水解羊骨粉的研究 被引量:5

摘要 为了进一步提高水解程度,以复合蛋白酶和风味蛋白酶双酶水解羊骨粉,比较同步水解和顺序水解效果,确定较优水解方式和条件。结果表明两种酶的顺序水解效果不及同步水解。双酶同步水解整体上优于单酶水解,且底物浓度9%、pH7.5时各指标均高于底物浓度12%、pH6时的水解效果。本研究确定的这两种酶水解羊骨粉的方式为同步水解,条件为:骨粉添加量9%、复合蛋白酶添加量5%(E/S)、风味蛋白酶添加量为4.25%(E/S)、调节起始pH值为7.5,50℃水解315min,此时的水解度为29.21%、氨基态氮含量为27.29%、多肽生成量为87.13mg·g-1、短肽得率为57.24%,其中多肽生成量和短肽得率极显著地高于单酶水解的67.75mg·g-1和43.39%(p<0.01)。 Protamex and flavorzyme were used sequentially or simultaneously to further improve the degree of hydrolyzing of sheep bone powder and to determine the better pattern and condition for hydrolyzing.Results suggested that simultaneous pattern was better than sequential pattern.9% bone powder at pH 7.5was better than 12% at pH 6.Hence the determined pattern for double-protease hydrolysis of sheep bone powder was simultaneous style and the condition was 9% substrate,5%(E/S)protamex,4.25%(E/S)flavorzyme,pH 6,50℃,315 min.Under this condition,the degreee of hydrolysis,amount of amino-acid nitrogen,reached to 29.21% and 27.29%.The amount of formed polypeptides and the obtaining rate of oligopeptides reached to 87.13 mg·g-1 and 57.24% respectively which is extremely higher than 67.75mg·g-1 and 43.39% of single-enzyme pattern(P〈0.01).
