钩端螺旋体糖脂蛋白对培养血管内皮细胞的粘附导致细胞内吞与病损 被引量:1

ENDOCYTOSIS AND CYTOTOXIN EFFECT INDUCED BY THE ATTACHMENT OF THE GLYOLIPOPROTEIN OF LEPTOSPIRES TO VASCULAR ENDOTHELIAL CELL IN CULTURE

摘要 作者提取不同群、型和毒力的构端螺旋体糖脂蛋白,以Rhodamine B荧光素和胶体金标记制备成探针,与体外培养的牛主动脉内皮细胞作用,以荧光显微镜和扫描电镜观察记录其过程,并以内皮细胞LDH、AcP漏出率和蛋白含量及细胞形态学作为细胞毒指标。结果表明,钧端螺旋体糖脂蛋白通过对培养血管内皮细胞的粘附,导致细胞内吞与病损。该过程可能属于一种特殊的受体介导内吞过程。 The Glycolipoprotein (GLP) of variousvirulent leptospires was extracted, purifiedand identified. The GLP was combined withfiuorescence rhodamine B and the colloidalgold as the probe. Meanwhile, the bovineaortic endothelial cell (EN) was isolated andcultured in the medium 199 'Nissui' . TheGLP-rhodamine B and GLP-colloidal goldprobe were added to cultured EN and incu-bated at 4℃ for 2 h then 37℃ for 2 h. Theinteraction between GLP-rhodamine B,GLP-CG probe and EN were observed andphotographied by fluorescence microscopy(Leitz) and scanning electromicroscopy. Thereleasing rate of lactic acid dehydrogenase(LDH), acid phosphatase (AcP) and proteinfrom the EN were detected. The results in-dicated that the GLP of Leptospira ihterro-gans serovar Lai, strain 017 attached to ENand a endocytosis was happened subsequent-ly. The LDH and AcP releasing rate weresignificantly higher than control and thecell protein was much less than control. Theresults showed that the GLP of L. interro-gans strain 017 had the cytotoxin effectthrough a specific attachment, endoeytosisin EN.The endocytosis probably is a recep-tor-mediated endocytosis.
