摘要
目的:探究桑葚花色苷对乳腺癌裸鼠肿瘤组织中VEGF、p53及Ki67表达的影响。方法:选择60只健康BALB/c裸鼠,按随机数字表分为5组,即对照组、桑葚花色苷低剂量组(SL组)、桑葚花色苷中剂量组(SM组)、桑葚花色苷高剂量组(SH组)以及阳性药(氟尿嘧啶)组,每组12只裸鼠。以注射乳腺癌MDA-MB-231细胞悬液到裸鼠右肩皮下的方法建立移植乳腺癌模型,分别检测和比较各组裸鼠的癌瘤湿重、癌重系数以及肿瘤组织中VEGF与p53、Ki67的表达。结果:治疗后,与对照组相比,各治疗组的癌瘤湿重水平及癌重系数均明显下降;与阳性药组相比,SM组及SH组的癌瘤湿重以及癌重系数下降程度更为明显,差异均具有统计学意义(P<0.05);治疗后,与对照组相比,各治疗组的VEGF与p53、Ki67阳性表达率均明显下降;且与阳性药组相比,SM组及SH组的VEGF与p53、Ki67阳性率下降程度更为明显,差异均具有统计学意义(P<0.05)。结论:桑葚花色苷能够明显抑制乳腺癌的发展,这可能与其抑制P53、Ki67在和VEGF的表达有关。
Objective: To explore the effect of mulberry anthocyanins on VEGF, p53 and Ki67 expressions in the cancer tissue of breast cancer-bearing nude mice. Methods: 60 healthy BALB/c nude mice were selected and divided into 5 groups according to random number table. The control group, mulberry anthocyanins low dose group(SL), mulberry anthocyanins dose group(SM), mulberry anthocyanins, high dose group(SH), as well as the positive drug group(fluorouracil), 12 nude mice in each group. Nude mice model of breast cancer were established by injection of MDA-MB-231 cells suspension to the right shoulder subcutaneously, the cancerous wet weight, weight coefficient of cancer and VEGF, p53 and Ki67 expresssions in the cancer tissue of breast cancer-bearing nude mice were detected and compared between different groups. Results: After treatment, compared with the control group, the cancerous wet weight and weight coefficient of cancer of treatment group were markedly reduced; compared with the positive drug group, the cancerous wet weight and cancerous weight coefficient of SM and SH group were declined more apparently(P〈0.05). After treatment, compared with the control group,the VEGF and p53, Ki67 positive expression rates of treatment group were significantly decreased; and compared with the positive drug group, the expressions of VEGF, p53 and Ki67 decreased more apparently,(P〈0.05). Conclusion: Mulberry anthocyanins could significantly inhibit the development of breast cancer, which may to correlated to the inhibition of P53 and Ki67 in and VEGF expressions.
