Expression of RSK4 mRNA in primary pancreatic carcinoma tissues and its clinicopathological significance

摘要 目的检测RSK4 mRNA在人原发性胰腺癌组织中的表达情况,探讨其临床病理诊断及预后意义。方法使用RT-PCR检测50例胰腺癌及30例癌旁组织中RSK4 mRNA的表达情况,并分析其表达与临床病理参数及术后生存时间的关系。结果 RSK4 mRNA在胰腺癌组织中相对表达量低于癌旁组织(P<0.05),ROC曲线下面积为0.695(95%CI 0.540-0.853,P=0.029);RSK4 mRNA在胰腺癌组织中的表达与远处转移情况有关(r=-0.364,P<0.05),而与其他临床病理参数无关(P>0.05);RSK4 mRNA阳性组中位生存时间19个月,阴性组中位生存时间8个月,阳性组预后明显优于阴性组(P<0.05);RSK4 mRNA、肿瘤分化程度和TNM临床分期是影响胰腺癌患者预后的因素,且均是独立的预后因素。结论 RSK4 mRNA可影响患者术后生存时间,其在胰腺癌的预后判断中可能具有一定的参考价值。 Purpose To investigate the expression and its clinicopathological and prognostic significance of RSK4 mRNA in human primary pancreatic carcinoma( PC). Methods Reverse transcription polymerase chain reaction( RT-PCR)was performed to detect the expression of RSK4 mRNA in fifty cases of PC and thirty cases of adjacent non-neoplastic pancreatic tissues. The relationship between RSK4 mRNA level and clinicopathological parameters of PC was analyzed by Chi-square test. Results The relative expression level of RSK4 mRNA in PC was significantly lower than adjacent nonneoplastic pancreatic tissues( P 〈0. 05). The expression of RSK4 mRNA in PC was reversely related to metastasis( r =- 0. 364,P 0. 05),however no statistically significant correlation was found between RSK4 mRNA expression and other clinicopathological parameters( P〉0. 05). The median survival time of the group of positive expression of RSK4 mRNA( 19 months) was longer than the negative one( 8 months,P〈 0. 05). The expression of RSK4,tumor differentiation as well as TNM clinical stage were the independent prognostic factors of PC. Conclusion The expression of RSK4 in PC can affect the survival of PC. It may be considered as a potential prognostic indicator.
