摘要
目的应用聚合酶链反应(PCR).变性梯度凝胶电泳(DGGE)技术,分析30对母子口腔唾液中菌群的相似性。方法选取3~5岁高龋、中龋和无龋儿童及其母亲各10对作为研究对象,取其非刺激性全唾液,分别提取细菌总DNA,进行16SrRNA的PCR扩增及DGGE分析。结果母子间口腔菌群组成具高度相似性。其中,无龋组母子间配对相似系数明显低于高龋组及中龋组(P〈0.05);高龋儿童的母亲平均龋失补牙面数(DMFS)明显高于中龋和无龋儿童的母亲(P〈0.05)。结论母亲是儿童口腔菌群获得的主要来源。
Objective This study aimed to analyze the similarity of the oral bacteria community in 30 mother-child dyads via polymerase chain reaction(PCR)-based denaturing gradient gel electrophoresis. Methods Unstimulated saliva samples were obtained from 30 mother-child pairs: 10 in caries-susceptive group, l0 in caries-moderate group, and l0 in caries-free group. DNAs were extracted for PCR amplification and denaturing gradient gel electrophoresis analysis. Results The compositions of the oral bacteria community in mother and child have a high degree of similarity. The oral bacteria community of children matched that of their mothers. The dice similarity coefficient of mother-child in the caries- free group was more statistically significant compared with the two other groups(P〈0.05). However, mothers of caries- susceptive children had significantly higher decay missing filling surface(DMFS) scores than mothers of caries-free and caries-moderate children(P〈0.05). Conclusion Children obtained oral bacteria from their mothers.
