摘要
目的探讨NDPK-A过表达对人膀胱癌细胞株T24增殖能力和超微结构的影响。方法通过脂质体转染和G418筛选,建立稳定表达外源NDPK-A的T24细胞株,免疫细胞化学法检测细胞内NDPK-A表达,MTT法测定细胞增殖能力,透射电镜观察细胞的线粒体超微形态。结果 NDPK-A过表达对T24细胞的增殖有抑制作用,引起了T24细胞内线粒体的固缩、内嵴变形,或发生肿胀和内部空洞化。结论 NDPK-A对膀胱癌细胞T24的线粒体有破坏作用,可能通过抑制细胞的新陈代谢而抑制其增殖。
Objective To investigate the effect of NDPK-A overexpression on the proliferation and ultrastructure of the T24 human bladder cancer cell line.Methods T24 cells were transfected by NDPK-A expressing plasmid with the aid of lipofectamine,followed by screening under the pressure of G418.Expression of NDPK-A protein was detected by immunocytochemistry.Cell proliferation was determined by MTT assay,and ultrastructure of mitochondria was observed under transmission electron microscope.Results Overexpression of NDPK-A not only caused significant inhibition of proliferation in T24 cells,but also led to pycnosis,swelling,cristae deformation and internal cavitation in mitochondria of T24 cells.Conclusion NDPK-A has detrimental effects on mitochondria in the T24 bladder cancer cells.Inhibitory effect on mitochondria metabolism might be one of the mechanisms through which NDPK-A suppresses proliferation of T24 cells.
