摘要
以叶上花为原料,微波提取水溶性多糖,用苯酚-浓硫酸法测定其多糖含量。研究不同提取溶剂、不同提取方法对叶上花多糖提取率的影响,通过单因素试验和正交试验对多糖提取工艺参数进行优化。结果表明,在所研究的提取溶剂和提取方法中,水为较佳提取溶剂,微波法为较佳提取方法。微波法提取叶上花多糖的最佳提取工艺参数为:料液比1∶40(g∶m L)、微波时间2.0 min、微波功率640 W。在此条件下,测得叶上花叶、茎多糖的平均提取率分别为22.82%、20.16%。该方法简便快速,重复性好,准确度较高,精密度良好,可用于叶上花多糖的提取。
The water soluble polysaccharides were extracted from Helwingia himalaica by microwave-assisted method, and the polysaccharide content was determined by phenol-sulfuric acid method.The effect of different extraction solvents and extraction methods on the extraction rate of polysaccharide was also studied.The parameters of extraction process of polysaccharide were optimized through single factor experiment and orthogonal experiment.The results showed that water was the best extraction solvent,and microwave was the best extraction method.The optimum extraction parameters of polysaccharide by microwave were as follows:ratio of solid to liquid was 1∶40(g∶mL),microwave time was 2.0 min, microwave power was 640 W.Under such conditions, the average extraction rates of polysaccharide in leaves and stems were 22.82% and 20.16% respectively.The method for extraction of polysaccharide from Helwingia himalaica had the advantages of simple operation, good reproducibility, high accuracy. Therefore,it could be used in the extraction and determination of the polysaccharide from Helwingia himalaica.
