摘要
利用超高效液相-二极管阵列检测器(UPLC-PDA)分析研究了造纸法再造烟叶各工艺环节产物中5种多酚(绿原酸、咖啡酸、莨菪亭、芸香苷及愈创木酚)的含量及其影响因素。结果表明:1再造烟叶原料中89.2%的多酚存在于碎烟叶中,烟梗中只有10.8%,其中原料中3种主要多酚(绿原酸,莨菪亭,芸香苷)占多酚总量的97.2%;2提取阶段,原料中90.3%的多酚转移到提取液中,二级提取固相物中多酚为9.7%,由于二级提取液相物的循环使用,最终提取液中多酚为97.1%;3真空浓缩阶段,绿原酸受热分解,损失量相当于多酚总量的10.0%,其余多酚变化不明显,最终涂布液中多酚占总量的89.7%;4打浆和抄造阶段,多酚随着水的流失而损失,片基中多酚剩余0.9%;5涂布阶段,多酚在有氧及高温下发生氧化,损失多酚48.7%,最终再造烟叶中多酚剩余41.9%。
Five polyphenols(chlorogenic acid,caffeic acid,scopoletin,rutin and guaiacol) generated from various paper-making reconstituted tobacco processes were analyzed by ultra performance liquid-diode array detector(UPLC-PDA).Impact of processing techniques on contents of polyphenols was also studied.Results showed that:1 89.2% of polyphenol compounds existed in crushed tobacco leaves and rest in stems.Three major polyphenols,namely chlorogenic acid,scopoletin and rutin,accounted for 97.2% of the total; 2 In extraction phase,90.3% of polyphenol compounds in raw material was transferred,and the rest 9.7% was in the secondary extraction solid phase and the final extraction had 97.1% of polyphenol due to the cycling use of secondary extract liquid phase.3 In vacuum concentration phase,about 10.0% chlorogenic acid was lost due to thermal decomposition,and the rest showed no obvious change.Polyphenols in final coating liquid accounted for about 89.7% of the total; 4 In beating and paper-making phase,polyphenols decreased sharply with loss of water and only about 0.9% polyphenols remained in base-sheet owing to water soluble polyphenol compounds; 5 48.7% of polyphenols was lost in coating phase due to oxidation and high temperature,and 41.9% of polyphenols remained in the final reconstituted tobacco.
