摘要
从稳定黄芩悬浮细胞系中筛选生长状况良好的黄芩悬浮细胞,研究了pH值、接种量和摇床转速对黄芩悬浮细胞中苯丙氨酸解氨酶(PAL)活性的影响.结果表明,在黄芩悬浮细胞生长初期,pH值为6.6时PAL活性略高,但纵观整个黄芩细胞生长周期,pH为5.8时的PAL活性明显更好;而当接种量为30 g·L-1,摇床转速为110 r·min-1时,黄芩悬浮细胞中的PAL活性亦是最佳.利用正交实验设计方案筛选出适于黄芩悬浮细胞PAL活性提高的培养条件:NH4+∶NO3为10∶40 mmol·L-1,蔗糖4%,6-BA 1.0 mg·L-1,2,4-D 0.2 mg·L-1.
Using the Scutellaria baicalensis suspension cells as the material, effects of pH, inoculum amounts and rotation rate on activities of phenylalanine ammomia lyase ( PAL ) from suspension ceils from Scutellaria baicalensis Georgi were researched. The results showed that the activities of PAL was the highest when pH was 6.6 during the the former days of the growth cycle of the Scutellaria suspension cells, however, we found that pH 5.8 was the higher on the whole of the growth cycle of the the suspension cells. Moreover, the activities of PAL in the suspension cells was also the highest when the amount of coverage for 30 g · L-1and the rotation rate of 110 r · min-1. On the other hand, the effects of different factors on the activities of PAL in suspension Scutellaria baicalensis Georgi ceils was studied by orthogonal design. The results showed that when 6-BA to 1 mg ·L-1, 2, 4-D to 0.2 mg · L-1, Sucrose to 4%, NH4+ : NO3- to 10 : 40, the activities of PAL was the highest.
