摘要
采用PDA-苯胺蓝显示法检测了20株木腐菌的产漆酶特性,初筛选出8株产漆酶菌株,通过液体发酵培养进一步检测其漆酶活性。结果显示,能够产生漆酶的菌株中只有6株检测到漆酶活性,其中SWFC9938产漆酶能力最强,最高平均酶活性可达1 656.82 U/m L,为高产漆酶菌株。Cu2+和藜芦醇诱导能显著提高SWFC9938的产酶能力,最高平均酶活性分别可达2 485.10和2 454.24 U/m L。藜芦醇能明显缩短最高产酶的发酵时间,发酵8 d时即出现酶活性高峰(对照出现在12 d以后),藜芦醇对SWFC9938菌株具有很好的诱导产酶活性。
Used PDA-aniline blue display method to detect the 20 strains producing laccase properties of wood rotting fungi,selected 8 strains producing laccase strains and developed further testing the laccase activity by liquid fermentation. The results showed that,the crude laccase which liquid fermentation among six potential strains exhibited high catalytic activity and the highest laccase activity approximated 1 656.82 U / mL for SWFC 9938. It was found that copper and veratryl alcohol were the best laccase inducers for strain SWFC 9938 with 2 485.10 U / mL and 2 454.24 U / mL respectively. Veratryl alcohol could obviously shorten the fermentation time of the maximum enzyme production,the maximumLaccase activity appeared at 8 days' cultivation(the maximumLaccase activity of control material was at 12 days' cultivation), indicating that veratryl alcohol might be laccase-induced potential.
